Radiolabeling and bioevaluation of dextran cysteamine mannose for sentinel lymph node detection

The aim of the present study was to radiolabel and bioevaluate the dextran cysteamine mannose (mw=22 kda) for sentinel lymph node detection. the radiolabeling was carriwd out by using isolink kit. various parameters were studied such as in vitro stability at room temperature up to 5h,protein binding and partition coefficient. bioevaluation was performed in rabbits; images were taken by using spect gamma camera,dissection was performed for studying the uptake of radiolabeled drug in popliteal/ iliac nodes and the extraction from popliteal node. percent injected dose per gram in various body organs was also studied. radiolabeling efficacy of the drug with radiometal was 96.3 ± 1%. 78.2 ± 3% of the radiolabeled drug showed binding with blood proteins. partition coefficient results showed that the radiopharmaceutical is hydrophilic in nature. in biodistribution study sentinel node was visualized within 15 min of post injection. drug shows 6.8 ± 2% uptake in popliteal node,0.68 ± 0.12% uptake in iliac node and 89.2 ± 5% popliteal extraction. the study demonstrates that radiolabeled dextran cysteamine mannose might prove a successful radiopharmaceutical for sentinel lymph node detection in future clinical applications.
