اثر تمرین هوازی همراه با مصرف عصاره پوست پسته بر بیان bax، کاسپاز 3 و bcl-2 بافت قلب موش‌های صحرایی تغذیه‌شده با غذای چرب

سابقه و هدف: رژیم غذایی چرب باعث افزایش آپوپتوز و بیماری‌های قلبی - عروقی می‌شود. بنابراین هدف از تحقیق حاضر، بررسی اثر 4 هفته تمرین هوازی همراه با مصرف عصاره پوست پسته بر بیان bax، کاسپاز 3 و bcl-2 بافت قلب موش‌های صحرایی تغذیه‌شده با غذای چرب بود.مواد و روش‌ها: این مطالعه از نوع تجربی است. موش‌های صحرایی ماده 12هفته‌ای به‌صورت تصادفی به 5 گروه 6تایی تقسیم شدند؛ کنترل مثبت (سالم)، کنترل منفی (چاق)، تمرین + چاقی، عصاره + چاقی، تمرین و عصاره + چاقی (تعامل تمرین و عصاره). تمرین 4 هفته با شدت متوسط 50-60% vo2max انجام شد. عصاره پوست پسته در دوز 60 میلی‌گرم به‌ازای هر کیلوگرم استفاده شد. برای بررسی بیان ژن‌ از روش qpcr استفاده شد.نتایج: نتایج نشان داد که بیان bax و کاسپاز 3 در گروه چاق بیشتر از گروه سالم بود (0/001=p). گروه‌های عصاره، ورزش و ورزش + عصاره در مقایسه با گروه چاق بیان bax و کاسپاز 3 بیشتری داشتند (0/001=p). بیان bcl-2 در گروه چاق و گروه ورزش کمتر از گروه سالم بود (0/001=p). همچنین بیان bcl-2 در گروه ورزش + عصاره بیشتر از گروه چاق بود (0/001=p). بین گروه ورزش و گروه ورزش + عصاره با گروه عصاره تفاوت معناداری وجود داشت.نتیجه‌گیری: رژیم غذایی پرچرب باعث افزایش bax و کاسپاز 3 و کاهش bcl-2 در موش‌های صحرایی می‌شود؛ با این‌حال ورزش و مصرف عصاره پوست سبز پسته به‌صورت توام باعث کاهش bax و کاسپاز 3 و افزایش bcl-2 می‌گردد.

the effect of aerobic exercise with pistachio skin extract on the expression of bax, caspase-3 and bcl-2 in heart tissue of fat-fed rats

background: fatty diet increases apoptosis and cardiovascular disease. therefore, this study aimed to evaluate the effect of 4-week aerobic exercise along with pistachio skin extract on the expression of bax, caspase-3 and bcl-2 in heart tissue of fat-fed rats.materials and methods: this is an experimental study. 12-week-old female rats were randomly divided into 5 groups of 6; positive control (healthy), negative control (obesity), exercise + obesity, extract + obesity, exercise and extract + obesity (interaction of exercise and extract). training was done for 4 weeks with moderate intensity of 50-60% vo2max. pistachio skin extract was used at a dose of 60 mg per kilogram. qpcr method was used to check gene expression.results: the results showed that the expression of bax and caspase 3 was higher in the obese group than in the healthy group (p=0.001). the extract groups, the exercise group and the exercise + extract group had more bax and caspase 3 expression compared to the obese group (p=0.001). the expression of bcl-2 in the obese group and the exercise group was lower than the healthy group (p=0.001). also, the expression of bcl-2 was higher in the exercise + extract group than in the obese group (p=0.001). there was a significant difference between the exercise group and the exercise+extract group with the extract group.conclusion: high-fat diet increase bax and caspase 3 and decreased bcl-2 in rats. however, exercise and consumption of green pistachio skin extract together decrease bax and caspase 3 and increased bcl-2.
