چکیده
|
سابقه و هدف: یکی از مکانیسمهای احتمالی برای محدودیت رشد سیستم عصبی و اختلالات رفتاری بهدنبال اعمال هیپوکسی، استرس اکسیداتیو است. با توجه به نقش آنتیاکسیدانی روغن ماهی، تحقیق حاضر با هدف بررسی اثر روغن ماهی بهطور همزمان با اعمال هیپوکسی مزمن بارداری بر حافظه، تغییرات مورفومتری مغز و استرس اکسیداتیو در زادههای بالغ موش صحرایی طراحی شد.مواد و روشها: در این آزمایش از موش صحرایی ماده باردار (نژاد ویستار) استفاده شد. موشها بهطور تصادفی به 5 گروه آزمایشی تقسیم شدند: کنترل، شاهد، هیپوکسی، روغن ماهی و گروه هیپوکسی تیمارشده با روغن ماهی. هیپوکسی با شدت اکسیژن 10 درصد و نیتروژن 90 درصد اعمال شد. مورفومتری مغز جنین در روزهای 15ام و 20ام بارداری و در زادههای 30 روزه سنجیده شد. حافظه احترازی غیرفعال و شاخصهای استرس اکسیداتیو در زادههای موش صحرایی 30 روزه ارزیابی شدند.نتایج: میانگین وزن مغز در جنینهای 15 و 20 روزه و زادههای 30 روزه گروه هیپوکسی بهطور معنیداری پایینتر از گروه شاهد بود (p<0/05 ). نتایج ارزیابی حافظه، بیانگر کاهش معنیدار حافظه احترازی در گروه هیپوکسی در مقایسه با گروه شاهد بود (p<0/05). استرس اکسیداتیو نیز در زادههای 30 روزه گروه هیپوکسی نسبت به گروه شاهد، افزایش معنیداری داشت و تیمار با روغن ماهی از کاهش این پارامترها جلوگیری کرد (p<0/05).نتیجهگیری: با توجه به نتایج بهدستآمده، تیمار روغن ماهی در طی هیپوکسی مزمن دوران بارداری، باعث کاهش محدودیت رشد مغز جنین، استرس اکسیداتیو و اختلال حافظه ایجادشده بهدنبال هیپوکسی شد.

Effect of fish oil treatment during chronic hypoxia in pregnancy on memory impairment, brain morphometry changes and oxidative stress in adult male rat offspring

Abstract
|
Background: One of the possible mechanisms to the nervous system growth restriction and behavioral impairment following hypoxia, is oxidative stress. According to the antioxidant effect of fish oil, the present study was designed to investigate the effect of fish oil treatment during chronic hypoxia in pregnancy on memory, brain morphometric changes and oxidative stress in adult rat offspring.Materials and Methods: In this study female pregnant rat (Wistar) were used; they were randomly divided into 5 experimental groups: control, sham, hypoxia, fish oil and hypoxia groups treated with fish oil. Hypoxia with 10% oxygen and 90% nitrogen intensity was applied. Fetal brain morphometry was examined on the 15th and 20th day of pregnancy and the 30dayold offspring. Passive avoidance memory and oxidative stress parameters were examined in the 30dayold rat offspring.Results: The brain weight average in the 15 and 20 dayold fetuses and 30dayold offspring of the hypoxia group was significantly lower than the sham group (P<0.05). Memory results evaluation indicated that the avoidance memory was significantly decreased in hypoxia group compared to the sham group (P<0.05). Oxidative stress also increased significantly in the 30dayold offspring of the hypoxia group compared to the sham group and fish oil treatment prevented the reduction of these parameters (P<0.05).Conclusion: According to these results, Fish oil treatment during chronic hypoxia in pregnancy can be attenuate fetal brain restriction, oxidative stress and memory impairment following hypoxia.
