Objective: the present study aim to investigate the relationship between neuronal autophagy and status epilepticus,and to probe the effects of chloroquine both on autophagy and epilepsy. method: 24 adult male wistar rats were randomly divided into three groups,named control group,pentamethazol induced epilepsy group and chloroquine pretreatment group. the behavior and electroencephalogram of rats were observed and recorded within 2h after injection. the nissl staining was used to detect the level of neuronal loss. and the levels of microtubule-associated protein light chain 3 (lc3) and p62 was reflected to the expression of autophagy. results: seizure intensity in the pentamethazol treatment group was more severe than that in the chloroquine pretreatment group and the control group (p<0.05). the number of survival neuron cells in the chloroquine pretreatment group was significantly increased compared to pentamethazol treatment group (p<0.05). the pentamethazol treatment group displayed a remarkable accumulation of lc3b-ii and a significant decrease of p62 when compared with control group (p<0.05). in the chloroquine pretreatment group,the level of lc3b-ii and p62 was notably higher than that in the pentamethazol treatment group (p<0.05). conclusion: results showed that pentamethazol injection notably increased the expression of autophagy and neuronal damage in the brain. results also suggested that chloroquine partially inhibited the hyperactivation of autophagy,played a beneficial role in neuroprotection in rats with pentamethazol-induced status epilepticus,and suppressed seizures to some extent. thus,our study indicated that chloroquine may be a potential antiepileptic drug.
