معرفی برخی شاخص‌های سرم، ایمنی و خون به عنوان شاخص سلامت در مولدین قزل آلای رنگین کمان (oncorhynchus mykiss) عاری از عوامل بیماریزای خاص

در این بررسی تلاش شده است با ارزیابی برخی از شاخص های خونی، سرمی و ایمنی به مبنایی جهت تعیین سلامت مولدین نسل پایه spf در مرکز نگهداری آنها دست یافت. به این منظور طی سه مرحله نمونه برداری قبل، همزمان و بعد از تکثیر از ماهیان مولد نگهداری شده در مرکز spf واقع در مرکز تحقیقات ماهیان سردآبی تنکابن و دو مزرعه منتخب (s و f) از تامین کنندگان مولد برای این مرکز، نمونه برداری شد. لازم به ذکر است که نمونه برداری در مرکز spf و مزارع منتخب از ماهیان متعلق به گله های مشترک انجام شد. در هر مرحله پس از بیهوشی و خونگیری از ماهیان میزان هماتوکریت، هموگلوبین، پروتئین تام، آلبومین، آنزمهای آلانین آمینوترانسفراز ( alt )، آسپارتات آمینو ترانسفراز (ast)، کراتین فسفوکیناز ( cpk )، آلکالین فسفاتاز (alp)، igm تام سرم و لیزوزیم اندازه گیری شد. نتایج نشان داد که قبل از فصل تکثیر میزان هموگلوبین، alt و igm در بین ماهیان سه مرکز تفاوت معنی داری نداشت ولی میزان لیزوزیم ماهیان مرکز spf بطور معنی داری از ماهیان مزرعه s و از نظر عددی از مزرعه f بیشتر بود. نتایج فصل تکثیر نشان داد میزان هماتوکریت، هموگلوبین، پروتئین تام سرم، آلبومین، igm تام سرم، ast، alt و میزان لیزوزیم در بین ماهیان سه مرکز فاقد تفاوت معنی داری بود. در حالیکه میزان alp ماهیان مرکز spf در مقایسه با مزرعه f تفاوت معنی دار داشت. پس از فصل تکثیر میزان هماتوکریت و وزن ماهیان گروه spf بطور معنی داری بیشتر از ماهیان دو مزرعه دیگر بود و هموگلوبین، igm و لیزوزیم بعضا بطور معنی دار و یا از نظر عددی از ماهیان دو مزرعه دیگر بیشتر بود. براساس نتایج حاصل از این بررسی به نظر میرسد استفاده از شاخص های خون و سرم می تواند به عنوان ابزاری جهت ارزیابی شرایط سلامت و نیز کیفیت شرایط نگهداری، مدیریت تغذیه، پرورش و بهداشت مورد استفاده قرار گیرد.

Introduction of some serum, immunity and blood indices as health indicators in rainbow trout (Oncorhynchus mykiss) brood stock of specific pathogen free fish (SPF)

In this study has been attempted to evaluate some hematological, serological and immunological indices to determine the baseline for determining the health of SPF brood stock. For this purpose, during three sampling stages, before, simultaneously and after breeding time, the blood samples were collected from brood stock fish at the SPF center in the Cold water fisheries research Center (Tonekabon) and two elected farms (F and S). The fish of SPF center were supplied of the two others farms (F and S) and sampling were conducted from same folk at the SPF center and elected farms. Hematocrit, hemoglobin, total protein, albumin, alanine aminotransferase (ALT), aspartate amino transferees (AST), certain phosphokinase (CPK), alkaline phosphatase (ALP), total IgM and lysozyme were measured. The results showed that before the breeding season, the levels of hemoglobin, ALT and total IgM were not significantly different among the fish in the three centers, but the lysozyme levels of SPF center were significantly and numerically higher than S and F fish farms respectively. Statistical analysis of data showed that there were no significant differences in hematocrit, hemoglobin, total protein, albumin, IgM, AST, ALT and lysozyme levels among the three groups during the breeding season. The results after the breeding season showed that the hematocrit and weight of SPF fish were significantly higher than those two other groups but hemoglobin, IgM and lysozyme levels were significantly and or numerically higher than those two other groups. Based on the results, it seems that the use of blood and serum indices can be used as a tool for health assessment condition, nutrition, rearing and hygiene managements. It seems that the health and nutrition management of the SPF center had been better than two other farms and it caused to improve the hemato serological and immunological parameters of the SPF fish in compared to the other elected farms fish after breeding time.
