   تجزیه فیلوژنتیک و هاپلوگروهی بزهای بومی ایران بر اساس توالی نوکلئوتیدی ناحیه Hvr1 ژنوم میتوکندری

چکیده    بزهای بومی ایران به عنوان یکی از ذخایر ژنتیکی ارزشمند محسوب می شوند و حفظ تنوع ژنتیکی آنها حائز اهمیت است. ژنوم میتوکندری در یک اکوتیپ و مقایسه آن با سایر اکوتیپ ها می تواند شاخص مناسبی از میزان تنوع موجود در آن جمعیت را ارائه دهد. هدف از پژوهش اخیر بررسی ساختار ژنتیکی و ترسیم روابط فیلوژنتیکی ناحیه hvr1 میتوکندری چهار توده جمعیتی بز های بومی ایران شامل سیستانی، پاکستانی، عدنی و سیاه لری (هر کدام 4 راس) و مقایسه آنها با توالی نوکلئوتیدی جایگاه مذکور در دیگر اکوتیپ های بز می باشد. استخراج dna کل به روش فنل کلروفرم انجام شد و واکنش تکثیر با استفاده از یک جفت آغازگر اختصاصی انجام گرفت. محصولات تکثیرشده پس از خالص‌سازی به روش سانگر تعیین توالی شدند. توالی های مذکور به همراه  توالی مشابه از ژنوم میتوکندری مربوط به سایر اکوتیپ های بز بدست آمده از مرکز ملی اطلاعات بیوتکنولوژی (ncbi) جهت تجزیه ژنتیکی و ترسیم درخت فیلوژنتیکی مورد استفاده قرار گرفت. با بررسی ناحیه hvr1 ژنوم میتوکندری در توده های مختلف بز در مطالعه حاضر 123 جایگاه چندشکلی و 16 هاپلوتیپ شناسایی شد. تجزیه واریانس مولکولی نشان داد که 15 درصد از تنوع بین جمعیت ها و 85 درصد در درون جمعیت ها است. کلیه توده های بز ایران استفاده شده در مطالعه حاضر در گروه هاپلوتیپی a که معمولاً در همه قاره ها یافت می شود گروه بندی شدند. مقادیر آماره های d و fs تست تاجیما (به ترتیب 2.14- و 9.55-) در مطالعه اخیر منفی بود که ممکن است به دلیل کوچک بودن اندازه نمونه مورد استفاده در این مطالعه باشد. نتایج این مطالعه نشان داد توالی نوکلئوتیدی ناحیه hvr1 میتوکندری ابزار مناسبی برای مطالعات ژنتیکی و گروه بندی توده های جمعیتی بز در ایران و دنیا است.

   Phylogenetic and Haplogroup analysis of native goats of Iran based on nucleotide sequence of HVR1 region of mitochondrial genome

Abstract    Iranian indigenous goats are considered as one of the most valuable genetic reserves and it is important to keep their genetic diversity. The mitochondrial genome in an ecotype and comparing it with other ecotypes can provide an appropriate mesures of the diversity of the population. The purpose of this study was to investigate the genetic structure and phylogenetic relationships analysis of mitochondria HVR1 in four populations of indigenous goats in Iran including of Sistani, Pakestani, adani and Black Lori goats (Each of them has 4 heads) and compare to the other species of goats in the world. Total DNA extraction was performed using phenolchloroform method and proliferation reaction was performed using a pair of special primers. Proliferation products were sequenced after purification by Sanger method. All of these sequences with17 sequences of the mitochondrial genome of other non Iranian goat ecotypes obtained from the National Center of Biotechnology Information (NCBI) were used for genetic analysis and phylogenetic tree mapping. Studying of HVR1 region of the mitochondrial genome in different goat populations, 123 polymorphic sites and 16 haplotypes were identified. The analysis of molecular variance showed that 15% of variation belongs to between populations and 85% within populations. All Iranian goat populations used in the present study were grouped in the Haplogroup group A, which were commonly found in world ecotypes. The values ​​of the D and Fs statistics of the Tajima test were negative (2.14 and 9.55 respectively) in the recent study, which may be due to the small size of the sample size. The results of this study showed that HVR1 region of the mitochondrial genome is a suitable tool for genetic studies and Haplogrouping of Iran and Non Iranian goat populations.
