Screening of SDS-degrading bacteria from car wash wastewater and study of the alkylsulfatase enzyme activity

Background and objectives: odum dodecy sufate (d) s one of the man surfactant components n detergents and cosmetcs,used n hgh amounts as a detergent n products such as shampoos,car wash soap and toothpaste. therefore,ts boremedaton by sutabe mcroorgansms s mportant. akysufatase s an enzyme that hydroyses sufate -ester bonds to gve norganc sufate and acoho. the purpose of ths study was to soate d-degradng bactera from tehran cty car wash wastewater,study bactera akysufatase enzyme actvty and dentfy the akysufatase enzyme codng gene. materials and methods: creenng of d-degradng bactera was carred out on basa sat medum contanng d as the soe source of carbon. amount of d degraded was assayed by methyene bue actve substance (mba). results and conclusion: identficaton of the sdsa gene was carred by pcr and subsequent sequencng of the 16 rdna gene and bochemca tests dentfied pseudomonas aeruginosa. ths bacterum s abe to degrade 84% of d after four days ncubaton. bactera soated from car wash wastewater were shown to carry the sdsa gene (670bp) and the akysufatase enzyme specfic actvty expressed from ths gene was determned to be 24.3 unt/mg. the resuts presented n ths research ndcate that pseudomonas aeruginosa s a sutabe canddate for d bodegradaton.
