مباحث اخلاقی ویرایش ژنوم و ژن‌درمانی با تاکید بر سیستم crispr-cas

ژن‌درمانی شامل ورود ژن‌های اختصاصی و عملکردی به درون سلول‌ها با هدف پیشگیری و درمان است. این فرایند ممکن است به‌صورت خارج بدنی (آزمایشگاهی) یا به‌صورت مستقیم از طریق داخل بدنی انجام شود. یکی از روش‌های نوین ژن‌درمانی و ویرایش ژنوم که در چند سال اخیر معرفی‌ شده و در حوزه‌ی مهندسی ژنتیک و زیست‌فناوری تحولات بزرگی ایجاد کرده، سیستم ویرایش ژنی crispr-cas است. این سیستم سریع‌تر و ارزان‌تر و دقیق‌تر از شیوه‌های رایج ویرایش ژن مانند روش‌های talenو zfns است. در ارتباط با سیستم ویرایش ژنی، همانند هر فرایند نوظهور دیگری، چالش‌های اخلاقی و نگرانی‌هایی مطرح شده است؛ برای مثال، می‌توان از به‌کارگیری احتمالی این تکنیک برای اهداف هنجارشکن مانند بیوتروریسم، جهش‌های زیان‌بار وارده به موجود زنده، به‌هم‌خوردن تعادل اکولوژیک نام برد. در مقاله‌ی مروری حاضر، از پایگاه داده‌های science direct، pubmed، sid، scopus و web of science استفاده و پس از انتخاب و مطالعه‌ی کامل مقالات مورد نیاز، اطلاعات لازم استخراج و خلاصه شد. با توجه به ملاحظات اخلاقی و حقوقی موجود در خصوص فرایندهای ژن‌درمانی، انتظار می‌رود شرایط مناسبی برای بهره‌گیری هرچه‌بهتر از ژن‌درمانی فراهم آمده و از نگرانی‌ها، لغزش‌های ممکن و سوءاستفاده‌های احتمالی کاسته شود. مقاله‌ی حاضر مباحث اخلاقی پیرامون ویرایش ژنوم و ژن‌درمانی را با تاکید بر سیستم crispr-cas شرح می‌دهد.

Ethical Issues in Genome Editing and Gene Therapy with Emphasizing on CRISPR-Cas System

Gene therapy entails the introduction of specific, functional genes into cells for therapeutic or preventive purposes. This process may be used in vitro or directly in vivo. One of the novel methods of gene therapy and genome editing that has been introduced in recent years and have made great changes in the field of genetic engineering and biotechnology is CRISPRCas geneediting system. This system is faster, cheaper, more accurate and efficient than another genome editing one, such as TALEN and ZFNs. As with any other emerging process, the ethical concerns and challenges associated with the genome editing methods including the potential application of this technique for nefarious purposes, such as, bioterrorism, harmful mutations to the living being, and disrupting ecological balance. This review was prepared using the databases of Science Direct, PubMed, SID, Scopus, Web of Science, and then, the selected papers were fully reviewed and information was extracted and summarized. According to the ethical and legal considerations surrounding the processes in gene therapy, it is expected that appropriate conditions will be provided to make the best use of gene therapy and to reduce concerns and possible abuses. The purpose of this review was to investigate ethical issues of the genome editing and gene therapy with an emphasis on the CRISPRCas system.
