تاثیر 6 هفته تمرین استقامتی همراه با مصرف عصاره هیدروالکلی گزنه بر میزان آپوپتوز بافت بیضه در موش‌های سفید بزرگ آزمایشگاهی دیابتی ناشی از استرپتوزوتوسین

مقدمه و هدف: دیابت نوعی بیماری متابولیکی است که با اختلالات ناباروری و آپوپتوز بافت بیضه همراه است. پژوهش حاضر با هدف بررسی تاثیر 6 هفته تمرین استقامتی همراه با مصرف عصاره هیدروالکلی گزنه بر آپوپتوز بافت بیضه موش‌‌های سفید بزرگ آزمایشگاهی دیابتی انجام گرفته است.مواد و روش ها: 58 سر موش سفید بزرگ آزمایشگاهی نر ویستار به‌طور تصادفی به پنج گروه «سالم کنترل»، «دیابت کنترل»، «دیابت تمرین»، «دیابت مکمل» و «دیابت تمرین مکمل» تقسیم شدند. با تزریق درون صفاقی محلول mg/kg 45stz حل‌شده در بافر سیترات تازه دیابت القا شد. عصاره گیاه گزنه به مدت 6 هفته به حیوان گاواژ شد. پروتکل تمرین استقامتی به مدت 6 هفته انجام شد و 48 ساعت پس از آخرین جلسه تمرینی، موش‌های سفید بزرگ آزمایشگاهی تشریح شدند و بافت بیضه‌شان استخراج شد. برای بررسی آپوپتوز بافت بیضه از روش رنگ‌آمیزی تانل استفاده شد. برای بررسی نتایج از آزمون‌های کلموگروف اسمیرنوف و لون برای بررسی نرمال‌بودن داده‌ها و همگنی واریانس‌ها استفاده شد. همچنین برای مقایسه گروه‌ها در متغیرهای مورد مطالعه از تحلیل واریانس یک‌سویه (آزمون تعقیبی توکی) استفاده شد.نتایج: آنالیز داده‌ها نشان داد که آپوپتوز در گروه‌های «دیابت تمرین»، «دیابت مکمل» و «دیابت تمرین مکمل» نسبت به گروه «دیابت کنترل» کاهش معناداری داشت (001/0 =p). عصاره گیاه گزنه و تمرین استقامتی در موش‌های سفید بزرگ آزمایشگاهی دیابتی موجب کاهش آپوپتوز بافت بیضه شدند.نتیجه‌گیری: مصرف عصاره هیدروالکلی گزنه به همراه تمرین استقامتی می‌تواند باعث کاهش آپوپتوز بافت بیضه موش‌های سفید بزرگ آزمایشگاهی دیابتی شود؛ لذا پیشنهاد می‌شود از ورزش به‌عنوان یک مداخله غیردارویی و گیاه گزنه که یک مداخله غیرشیمیایی است در جهت کاهش عوارض ناشی از دیابت در حیطه باروری استفاده شود.

the effect of 6-week endurance training with the consumption of nettle hydroalcoholic extract on testicular tissue apoptosis in streptozotocin-induced diabetic rats

background and objective: diabetes is a metabolic disease that is associated with infertility disorders and testicular tissue apoptosis. the present study was conducted with the aim of investigating the effect of 6 weeks of endurance training along with the consumption of nettle hydroalcoholic extract on the apoptosis of the testicular tissue of diabetic rats.materials and methods: 58 male wistar rats were randomly divided into five groups: &healthy control&, &control diabetes&, &exercise diabetes&, &supplementary diabetes& and &supplementary exercise diabetes&. diabetes was induced by intraperitoneal injection of 45 mg/kg stz dissolved in fresh citrate buffer. nettle plant extract was fed to the animals for 6 weeks. the endurance training protocol was performed for 6 weeks and 48 hours after the last training session, the rats were dissected and their testicular tissue was extracted. tunnel staining method was used to check the apoptosis of testicular tissue. the results of the kolmogorov smirnov and lunn tests were used to check the normality of the data and the homogeneity of the variances. also, one-way analysis of variance (tukey’s post hoc test) was used to compare groups for the studied variables.results: data analysis showed that apoptosis was significantly reduced in the &exercise diabetes&, &supplementary diabetes& and &supplementary exercise diabetes& groups compared to the &control diabetes& group (p = 0.001). nettle plant extract and endurance exercise in diabetic rats reduced testicular tissue apoptosis.conclusion: consumption of nettle hydroalcoholic extract along with endurance exercise can reduce the apoptosis of testicular tissue in diabetic rats. therefore, it is suggested to have exercise as a non-pharmacological intervention and nettle plant, which is a non-chemical intervention, to reduce the complications caused by diabetes in the field of fertility.
