ارزیابی فعالیت سمیت سلولی، مطالعات برهمکنش با Dna و آنزیم تیوردوکسینردوکتاز کمپلکس های ناجورهسته دوفلزی پلاتین(Ii) -طلا (I) به عنوان عوامل بالقوه ضدسرطان

چکیده
|
زمینه و هدف: دارو های شیمی درمانی بر پایه ی فلزات پلاتین مانند داروی سیسپلاتین امروزه بطور وسیع در درمان سرطان مورد استفاده قرار میگیرند. اما به دلیل عوارض جانبی زیاد استفاده از این ترکیبات محدود شده است. تمام تلاشهای امروزه برای طراحی داروهای جدید پلاتینی و همچنین داروهای هیبریدی از طلا و پلاتین میباشد که پایه عملکرد آنها مشابه سیسپلاتین است ولی دارای عوارض جانبی کمتر و طیف اثر بیشتر روی انواع سرطان باشند. روش بررسی: در این مطالعه، فعالیت سمیت سلولی کمپلکسهای ناجورهسته دو فلزی پلاتین(ii)طلا(i) بر روی ردههای سلولی سرطانی دهانه رحم (hela)، کولون (sw1116)، پستان (mda-mb-231) و همچنین سالم پستان (mcf-12a) به روش mtt مورد بررسی قرار گرفت. مطالعات داکینگ مولکولی این ترکیبات با dna و تیوردوکسینردوکتاز (trxr) به عنوان اهداف ترکیبات پلاتین و طلا توسط نرم افزارautodock 4.2 انجام گردید. با استفاده از روش ستاره دنبالهدار فعالیت ژنوتوکسیسیتی بهترین ترکیب سیتوتوکسیک نیز مورد ارزیابی قرار گرفت. یافته ها: در ارزیابی فعالیت سیتوتوکسیک، کمپلکسهای 1b و 2b اثرات سیتوتوکسیک بهتری نسبت به سیسپلاتین از خود نشان دادند. تمامی ترکیبات اثر سمیت پایین تری نسبت به سیسپلاتین روی رده سالم پستان (mcf-12a) نشان دادند. برهمکنش ترکیبات با dna و trxr توسط داکینگ نیز بررسی گردید و نحوه و چگونگی اتصال ترکیبات به آنها مشخص گردید. همچنین اثر این ترکیبات بر واپاشی dna با روش ستاره دنباله دار مورد تایید قرار گرفت. نتیجه گیری: کمپلکسهای 1b و 2b این قابلیت را دارند تا با بررسی های بیولوژیک بیشتر، در درمان سرطان نقش موثری را ایفا کنند.

Abstract
|
Background and Aim: Platinumbased chemotherapy drugs like cisplatin are widely used in the treatment of cancer. However, because of its high side effects, the use of these compounds is limited. All of today’s efforts is to design new platinum drugs, as well as hybrid of gold and platinum, which have a similar profile with cisplatin but have less side effects and wide spectrum on different cancer types. Material and Method: In this study, the cytotoxic activity of heterobimetallic Pt(II)Au(I) complexes on the cancer cell lines such as cervix (Hela), colon (SW1116), breast (MDAMB231) as well as normal breast (MCF12A) cell line was evaluated using MTT method. Molecular docking studies of these compounds with DNA and thioredoxin reductase (TrxR), as targets for platinum and gold compounds, were conducted using AutoDock 4.2 software. Using the comet assay, genotoxicity of the best cytotoxic compound was also evaluated. Result: In evaluating the cytotoxic activity, 1b and 2b complexes exhibited greater cytotoxicity than cisplatin. All compounds exhibited lower toxicity than cisplatin on normal breast cell line (MCF12A). The interaction of complexes with DNA and TrxR using molecular docking was also investigated and their binding mode and binding site were determined. The genotoxicity effect of these compounds was also confirmed by the comet assay. Conclusion: These complexes with further biological studies, have the ability to play a more effective role in the treatment of cancer.
