اثر تکاملی محرومیت از نور بر القای تقویت درازمدت (Ltp) در نورونهای ناحیهی Ca1 هیپوکامپ موش صحرایی

چکیده
|
مقدمه: علاوه بر فعالیت ذاتی نورونها، سیگنالهای حسی در دوران بحرانی تکامل مغز اثر شگرفی در ساختار و عملکرد مغز دارند. در این مطالعه اثر تکاملی محرومیت از نور بر روی پدیدهی تقویت پاسخهای میدانی ثبت شده از ناحیهی ca1 هیپوکامپ مورد بررسی قرار گرفته است.روشها: در این مطالعهی تجربی، 48 سر موش صحرایی نر در دو گروه روشنایی (lr یا light reared) و تاریکی (dr یا dark reared) قرار گرفتند. حیوانات lr از بدو تولد تا لحظهی آزمایش در شرایط معمول حیوانخانه و موشهای dr در شرایط تاریکی کامل قرار گرفتند. سپس حیوانات هر گروه به 3 زیر گروه 2، 6 و 10 هفته (هر گروه 8 سر) تقسیم شدند. با تحریک مسیر پریفورنت، پاسخهای میدانی نورونها در ناحیهی ca1 برای مدت 30 دقیقه ثبت گردید. سپس تحریک تتانیک اعمال شد و آزمایش به مدت 120 دقیقهی دیگر ادامه یافت.یافتهها: همزمان با افزایش سن، از اندازهی پاسخهای پایهی حیوانات lr کاسته و بر اندازهی پاسخهای حیوانات dr افزوده شد. همچنین، اگرچه محرومیت از بینایی مانع از القای تقویت درازمدت (ltp یا long-term potentiation) در ناحیهی ca1 هیپوکامپ نشد، اما از میزان تقویت آن کاست و با افزایش سن نیز در هر دو گروه حیوانات از اندازهی پاسخ بعد از القای ltp کاسته شد.نتیجهگیری: به نظر میرسد که تغییر در تجربهی بینایی از طریق تغییر در فعالیت نورترانسمیترهای مغزی باعث ایجاد اختلال در فعالیت نورونهای ناحیهی ca1 هیپوکامپ قبل و بعد از القای ltp میشود.

Developmental Effect of Dark Rearing on LongTerm Potentiation (LTP) Induction in CA1 Area of Hippocampus

Abstract
|
<p><strong>Background:</strong> In the critical period of brain development, in addition to natural function of neurons, the environmental signals strikingly affect the brain structure and function. In this study, the developmental effect of dark rearing on induction of longterm potentiation (LTP) in responses of neurons of CA1 area of hippocampus was evaluated.<strong></strong></p><p><strong>Methods:</strong> This experimental study was carried out on 2 groups of male rats kept in a standard 12hour light/dark condition (Light reared or LR) or a in complete darkness (Dark reared or DR) since birth through the study. Each group, in turn, was divided into 3 groups of 2, 6 and 10 weeks old subgroups (n = 8 for each). Stimulating the perforant path, field potentials were recorded in the CA1 area for 30 minutes. Then, the tetanic stimulation was applied to the Schaffer collaterals and the field potentials were pooled for 120 minutes posttetanus.<strong></strong></p><p><strong>Findings:</strong> While the response amplitude of the basic responses demonstrated an agedependent decrease in the LR animals, it showed a substantial increase in the DR ones. Dark rearing declined the degree of potentiation in the tetanized responses; however, the posttetanus recordings showed a significant LTP yet. The LTP induction was reduced in either LR or DR animals.</p><p><strong>Conclusion:</strong> It seems that the visual experience modifications weaken both basic responses and LTP induction in neurons of CA1 area of hippocampus.<strong></strong></p>
