چکیده
|
مقدمه: شیمیدرمانی یکی از پرکاربردترین روشهای درمان سرطانها میباشد. مهمترین مرحلهی درمان، انتخاب موثرترین دارو برای ایجاد بیشترین اثر سمیت بر سلولهای سرطانی است. دوستاکسول دارویی است که هنگام استفادهی ترکیبی با رادیوتراپی میتواند اثرات درمانی چشمگیری نظیر متوقف کردن چرخهی سلولی در فاز g_2/m، که مرحلهای بسیار آسیبپذیر به تابشهای یونیزان است، داشته باشد. مدت زمان اثردهی دارو با سلول یکی از عواملی است که اثر آن تا کنون در مطالعات سلولی این دارو به عنوان یک عامل متغیر مورد بررسی قرار نگرفته است. در این مطالعهی تجربی، ما به بررسی تاثیر تفاوت زمان اثردهی داروی دوستاکسول در افزایش میزان مرگ سلولهای سرطانی پستان ردهی 7-mcf در محیط برونتنی پرداختیم. روشها: در این مطالعه، سلولها را به سه گروه شاهد، 1 با مدت زمان اثردهی 5 ساعت و 2 با مدت زمان اثردهی 24 ساعت در غلظتهای مختلف داروی دوستاکسل تقسیم کردیم. میزان حیات سلولی از طریق آزمون mtt [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] در سه بازهی زمانی 24، 48 و 72 ساعت بعد از آزمایش سنجیده شد. یافتهها: میزان مرگ سلولی، وابسته به غلظت دارو بود. در مقایسه بین دو گروه 1 و 2، کاهش معنیداری در حیات سلولی، به ویژه در غلظتهای بالا مشاهده شد که نشاندهندهی تاثیر مدت زمان اثردهی بود. نتیجهگیری: مطالعهی ما اثبات کرد که مدت زمان اثردهی داروی دوستاکسول عاملی مهم در میزان مرگ سلولی است و هنگام کاربرد همزمان دوستاکسل با رادیوتراپی در مطالعات سلولی، باید به عنوان یک عامل متغیر مورد نوجه قرار گیرد.

Abstract
|
<div><p><strong>Background:</strong><em> </em>Over the past decade, researchers have used the existing knowledge about pathways messaging protocols to successfully stimulate stem cells to generate neurons. Due to the ease of access to adipose tissueobtained stem cells rather than other sources, whereas estrogen factor could be used to improve neural differentiation, antilogous transplantation of differentiated neurons would be widely used for certain degenerative neurological diseases such as Parkinson's disease and spinal cord injuries. The purpose of this study was to evaluate the effect of estrogen on expression of microtubuleassociated protein2 (MAP2),<em> </em>glial fibrillary acidic protein (GFAP)<em> </em>and Nestin markers.</p><p><strong>Methods:</strong><strong> </strong>After the isolation of stem cells from adipose tissue, the neural induction was carried out through neurosphere construction; then, final differentiation of the cells was performed. Neurospheresinged cell were transferred to neural induction medium (control group). In the estrogentreated group, estrogen was added to the culture medium until the end of the day of distinction. Then, evaluation of the expression of neural markers, was performed using reverse transcriptionpolymerase chain reaction (RTPCR) technique. In addition, MTT assay [3(4,5dimethylthiazol2yl)2,5diphenyltetrazolium bromide] was performed to assess cell viability.</p><p><strong>Findings:</strong><strong> </strong>The mean expression of GFAP and Nestin markers were down regulated in treated group compared to controls (P < 0.05). The difference between the mean of MAP2 expression was not significant between the two groups. In addition, the difference between the mean of cell viability was not significant between two groups, too.</p><p><strong>Conclusion:</strong> In this study, we found that estrogen can decrease the expression of neuronal markers. However, to determine the effect of estrogen on neurogenic differentiation of stem cells, next studies should be done broader using other precise techniques.</p></div><strong>Keywords:</strong> Estrogen, Neurogenesis, Adiposederived stem cell, Reverse transcriptionpolymerase chain reaction (RTPCR)
