بررسی بیان گیرنده انسانی فاکتور رشد اپیدرمی 2 (Her2) در سلول‌های سرطانی افراد مبتلا به سرطان روده بزرگ

زمینه و هدف: سرطان روده چهارمین سرطان شایع در دنیا می باشد. تعداد موارد جدید بیماری از سال 1975 در حال افزایش است. فعالیت بیش از اندازه خانواده egfr در انواع سرطان ها گزارش شده است و این امر نشان دهنده نقش گیرنده انسانی فاکتور رشد اپیدرمی 2 (her2) به عنوان یکی از اعضای این خانواده در بسیاری از سرطان ها به اثبات رسیده است. هدف از این مطالعه بررسی میزان بیان her2 در سرطان روده و ارتباط احتمالی آن با پیشرفت مراحل بیماری می باشد.روش کار: در این مطالعه 30 نمونه بافت بلوک پارافینه ی افراد مبتلا به سرطان روده و 10 نمونه غیر توموری مورد بررسی قرار گرفت. rna نمونه ها، پس از پارافین زدایی با محلول rnx استخراج و سپس سنتز cdna بر روی نمونه ها انجام شد. در انتها میزان بیان ژن her2 به کمک تکنیک realtime pcr نسبی تعیین گردید.یافته ها: نتایج نشان داد که بیان نسبی ژن her2 در افراد بیمار به صورت میانگین نسبت به نرمال به میزان 7.48 برابر افزایش داشته است. مقایسه میزان بیان در نمونه ها نشان دهنده افزایش بیان her2 در بیماران در مرحله iii نسبت به نمونه های در مرحله ii و i بود. همچنین ارتباط معناداری با افزایش سن، مشاهده گردید که این ژن در افراد بالای 50 سال بیشتر از افراد زیر 50 سال بیان می گردد.نتیجه گیری: با توجه به افزایش میزان بیان her2 در نمونه های سرطان روده بزرگ می توان بیان نمود که ارزیابی میزان بیان her2 می تواند هدفی احتمالی در تشخیص و پیش آگهی، سرطان روده در کنار سایر فاکتورها مطرح باشد.

Assessment of the expression level of (HER2) human epidermal growth factor receptor 2 in tumor cells derived from individuals with colorectal cancer

Background: Colorectal cancer is the fourth most common cancer in the world. It is the third largest risk factor for cancer individuals. The number of new cases is rising since 1975. Hyper activities of EGFR family have been reported in various cancers. The purpose of this study was to investigate the expression levels of HER2 in colorectal cancer and its possible association with advanced stages of disease.Methods: In this study 20 samples of colorectal cancer paraffin block and 10 nontumor individual samples were collected. After deparaffin, RNA molecules were extracted from samples by RNX plus solution, after that CDNA was synthetized by reverse transcription method and finally the relative HER2 gene expression was evaluated by Real Time PCR.Results: In 20 cases of colorectal cancer compared with nontumor samples, over expression of HER2 gene was observed. The increased expression compared with nontumor samples observed in all patient samples and the amount of over expressed HER2 in patients with stage III was more than stage II and I. expression levels were also increased by increasing the age. The observation showed that the increased expression of HER2 occurs from the early stages of the disease.Conclusion: Due to the increased HER2 expression in colorectal cancer, it is clear that the HER2 assessment can be considered as a valuable prognostic factor for screening especially in early stages of disease.
