القای سمیت سلولی و آپوپتوز توسط عصاره هیدرو الکلی کبابه چینی در سلول‌های سرطانی پستان

پیش‌زمینه و هدف: سرطان پستان به‌عنوان یکی از شایع‌ترین انواع سرطان، همچنان تهدیدی جدی برای سلامت زنان در سراسر جهان محسوب می‌شود. به علت عوارض جانبی بالای شیمی‌درمانی، جستجو برای ترکیبات طبیعی با خاصیت ضد سرطانی و عوارض جانبی کمتر، از اهمیت بالایی برخوردار است. هدف از این مطالعه، بررسی سمیت سلولی عصاره هیدرو الکلی کبابه چینی روی سلول‌های سرطانی پستان بود.مواد و روش کار: در این مطالعه، سلول‌های سرطانی mcf-7 در معرض غلظت‌های مختلف عصاره هیدرو الکلی کبابه چینی قرار گرفتند. سپس، با استفاده از آزمون mtt، بقای سلولی سنجیده شد. همچنین، تغییرات مورفولوژیکی سلول‌ها و وقوع آپوپتوز با استفاده از رنگ‌آمیزی‌های گیمسا و اتیدیوم بروماید-آکریدین اورنج بررسی گردید.یافته‌ها: نتایج حاصل نشان داد که عصاره به‌صورت وابسته به غلظت، باعث کاهش بقای سلولی در سلول‌های سرطانی mcf-7 شد. ic50 عصاره در این مطالعه mg/ml 1 تعیین گردید. همچنین، مشاهده تغییرات مورفولوژیکی مشخص در سلول‌های تیمار شده با عصاره، نشان‌دهنده القای مرگ سلولی در سلول‌ها از نوع آپوپتوز بود.بحث و نتیجه‌گیری: یافته‌های این مطالعه نشان داد که عصاره هیدرو الکلی گیاه کبابه چینی دارای اثرات کشندگی قابل‌توجه بر روی سلول‌های سرطانی پستان mcf-7 بود. این یافته‌ها، می‌تواند پتانسیل بالای این گیاه را به‌عنوان یک منبع طبیعی برای توسعه داروهای جدید ضد سرطان نشان دهد.

induction of cytotoxicity and apoptosis by hydroalcoholic extract of piper cubeba l in breast cancer cells

background & aims: breast cancer, as one of the most common types of cancer, remains a significant threat to women’s health worldwide. due to the high incidence of side effects associated with chemotherapy, the search for natural compounds with anticancer properties and fewer side effects is of great importance. the aim of this study was to investigate the cytotoxic effect of the hydroalcoholic extract of piper cubeba l. on breast cancer cells.materials & methods: in this study, mcf-7 cancer cells were exposed to different concentrations of the hydroalcoholic extract of piper cubeba l. subsequently, cell viability was measured using the mtt assay. additionally, morphological changes in cells and the occurrence of apoptosis were investigated using giemsa staining and ethidium bromide-acridine orange staining methods.results: the results showed that the extract, in a concentration-dependent manner, reduced cell viability and induced apoptosis in mcf-7 cancer cells. a concentration of 1 mg/ml was determined as the ic50 of the extract in this study. moreover, the observation of distinct morphological changes in cells treated with the extract indicated the induction of apoptotic cell death.conclusion: the findings of this study demonstrate that the hydroalcoholic extract of piper cubeba l. has significant cytotoxic effects on mcf-7 breast cancer cells. these findings indicate the strong potential of this plant as a natural source for the development of new anticancer drugs.
