Parkinson’s disease is a neurodegenerative disorder characterized by behavioral symptoms in animals, such as reduced interaction with the environment, repetitive behaviors, separation anxiety, and heightened fear responses to novel stimuli. although parkinson-like conditions are rare in pets, the contribution of dopaminergic neuron loss and oxidative stress to the disease process exhibits mechanisms similar to those observed in humans. structurally, the disease is marked by neuronal loss and α-synuclein aggregates in the substantia nigra, leading to striatal dopamine deficiency. 6-hydroxydopamine (6-ohda), an oxidizable dopamine analog taken up via the dopamine transporter and exhibiting selective toxicity toward dopaminergic neurons in the substantia nigra pars compacta (snpc), is widely used to develop in vitro models of parkinson’s disease. previous studies have reported the potential of small-molecule phytocomponents to ameliorate degenerative changes in brain tissues. calligonum polygonoides, an endemic, perennial plant resistant to harsh environmental conditions, possesses antioxidant, antimicrobial, and cytoprotective properties. in this context, the present study investigated the neuroprotective effects of the methanol extract of calligonum polygonoides (msecp) in a 6-ohda-induced in vitro parkinson’s model.to establish a parkinson s model in cell culture, sh-sy5y (neuroblastoma) cells were exposed to 200 µm 6-ohda for 15 minutes. subsequently, cells were treated with calligonum polygonoides extract (1 µg/ml, 5 µg/ml, and 10 µg/ml). after 24 hours, cell viability was assessed using the mtt assay. oxidative stress parameters were estimated via total antioxidant capacity (tac), total oxidant status (tos), and klotho levels.treatment with 6-ohda was found to increase apoptosis in sh-sy5y cells by approximately 55% compared to the control group. tac and tos analyses indicated that this increase was associated with oxidative stress. calligonum polygonoides extract was found to reduce 6-ohda-induced apoptosis and increase cell viability in a dose-dependent manner. furthermore, the observed klotho levels supported these findings.this study demonstrated the neuroprotective effects of calligonum polygonoides extract in an in vitro 6-ohda-induced parkinson s disease model using mtt, tac, tos, and klotho assays in sh-sy5y cells. to gain deeper insights into the underlying mechanisms, additional investigations employing in vivo parkinson’s models are warranted.
