   the effect of stachys lavandulifolia on prostate cancer cell line (lncap)

چکیده    Prostate cancer is one of the leading causes of cancer-related deaths among men, and despite advances in treatment, challenges such as drug resistance and recurrence remain significant stachys lavandulifolia, a medicinal plant known for its bioactive compounds, has demonstrated potential in various biological activities, including anticancer effects. this study aims to evaluate the cytotoxicity and anticancer potential of stachys lavandulifolia on the lncap prostate cancer cell line. the cytotoxic effect of stachys lavandulifolia was assessed using the mtt assay, which determined the ic50 value to be 1.09 mg/ml. five different treatment groups were established with varying concentrations of the plant extract: 0.125, 0.25, 0.5, 1, and 2 mg/ml. the cells were seeded in 96-well plates, and after 48 hours of exposure to the extract, the mtt assay was performed to assess cell viability. the ic50 value was calculated based on the concentration that inhibited 50% of cell viability. subsequently, flow cytometry evaluated apoptosis induction, revealing that a significant percentage of lncap cells underwent apoptosis following treatment with the plant extract at the ic50 concentration (1.09 mg/ml). flow cytometry analysis was performed using annexin v-fitc and propidium iodide (pi) staining, and the percentage of apoptotic cells was calculated based on the data. additionally, acridine orange staining was performed to confirm apoptosis induction further. these findings suggest that stachys lavandulifolia exhibits promising anticancer activity against prostate cancer cells by inhibiting cell proliferation and promoting apoptosis.
