Abstract
stachys lavandulifolia is a phytochemically rich medicinal plant containing diverse bioactivecompounds and photosynthetic pigments—particularly chlorophylls—associated with antioxidant andtherapeutic properties. this study investigates the efficiency of three common solvents (distilled water, methanol, and ethanol) in extracting chlorophyll pigments and related constituents from the driedaerial parts of s. lavandulifolia collected from the highlands of kerman. the extraction process wasconducted through maceration at room temperature. solvent recovery analysis showed that methanol(85%) and ethanol (82%) exhibited superior physical extraction efficiency compared to distilled water, mainly due to reduced interaction with cellulosic tissues and lower swelling effects. to determinepigment concentration, a dual-method approach was employed: ai-assisted image analysis (rgb/hsvcolor profiling) to quantify green intensity, validated by spectrophotometric measurements based onarnon’s equations. the results revealed that alcoholic solvents—especially ethanol—produced darkgreen extracts with maximum absorbance at 645 and 663 nm, indicating higher extraction yields forchlorophyll a and b. in contrast, distilled water, due to its strong polarity and inability to dissolvenonpolar pigments, produced a yellow extract devoid of chlorophyll. overall, ethanol is identified asthe optimal solvent for simultaneous extraction of lipophilic pigments and active phytochemicalcompounds. additionally, this study presents an innovative, low-cost, and engaging model for teachingthe concept of “solvent polarity” in 10th-grade chemistry education
