   in vitro regeneration of eruca sativa mill shoots

چکیده    Different tissue culture techniques were studied. there is limited information about the in-vitro propagation of eruca sativa mill. this experiment was conducted as factorial in a completely randomized design with three replications in two separate experiments with different hormonal treatments. the first experiment with different concentrations of bap control, 0.5, 1, 1.5 and 2 mg/l in combination with naa at concentrations control, 0.5, 1, 1.5 and 2 mg/l and the second experiment of bap with concentrations control, 0.5, 1, 1.5 and 2 mg/l in combination with 2,4-d with concentrations control, 0.5, 1, 1.5 and 2 mg/l and interaction of naa with bap was used. significant differences were observed between treatments at the level of 0.05 at the interaction between naa, bap and stem micro sample. in first experiment, the highest number of regeneration was observed in stem micro sample and 2 mg/l naa and 1 mg bap as well as leaf micro sample and 1.5 mg/l naa and 1.5 mg/l bap. in the second experiment, interaction effects were significant for all traits at the 0.01 level, except for interaction of bap at micro sample and interaction of bap at 2,4-d for regeneration number which was significant at 0.05 level. with using growth regulators, for the most callus formation, stem micro sample growth was suggested as the best micro sample. as well as the stimulatory role of 2.4-d in vitreous in eruca sativa callus formation. high 2.4-d increases callus formation but low concentration reduces callus formation. results of this experiment showed that the main effect of micro sample and bap on number of regeneration was not significant. but the main effect of 2,4-d on number of regeneration was significant (p≤0.01), and the highest number of regeneration was obtained at concentrations of 0.5 and 1.5 mg/l 2,4-d.
