مقایسۀ روش‌های مختلف استخراج ملانین طبیعی از پشم خودرنگ

در این تحقیق ملانین طبیعی به سه روش متفاوت از پشم خودرنگ استخراج شد. در روش اول از سدیم هیدروکسید، در روش دوم از هیدروکلریک اسید و در روش سوم از مایع یونی تترابوتیل­آمونیوم هیدروکسید استفاده شد. به منظور شناسایی ملانین و مقایسه سه روش استخراج از اسپکتروفتومتر فرابنفش-مرئی و طیف‌سنج مادون قرمز استفاده شد و بازدهی وزنی فرآیند استخراج محاسبه شد. نتایج نشان داد تترابوتیل­آمونیوم هیدروکسید، بیشترین مقدار ملانین را استخراج می‌کند. از طرفی طیف جذبی فرابنفش-مرئی نشان داد ملانین استخراج شده با هیدروکلریک اسید بیشترین جذب را داشته و در نتیجه برای کاربرد محافظت در برابر پرتو فرابنفش مناسب است. در مقابل ملانین استخراج شده با تترابوتیل­آمونیوم هیدروکسید با توجه به قله اصلی باریک و نبود قله فرعی از بیشترین خلوص برخوردار است. با توجه به طیف مادون قرمز گروه‌های عاملی ملانین در پودرهای استخراج شده شناسایی شدند.

comparison of different methods of extracting natural melanin

in this research, natural melanin was extracted from pigmented wool using three different methods. sodium hydroxide, hydrochloric acid, and tetrabutylammonium hydroxide ion liquid were used in the first, second, and third methods, respectively. the ultraviolet-visible spectrophotometer and infrared spectrometer were utilized to identify melanin and compare three extraction methods, and then the weight efficiency of the extraction process was calculated. according to the results, tetrabutylammonium hydroxide extracted the largest amount of melanin. on the other hand, the ultraviolet-visible absorption spectrum indicated that melanin extracted with hydrochloric acid had the highest absorbance; hence, it was suitable for protection against ultraviolet rays. on the contrary, melanin extracted with tetrabutylammonium hydroxide had the highest purity due to a narrow main peak and the absence of secondary peaks. melanin functional groups were detected in extracted powder according to the infrared spectrum
