مروری بر روش های استخراج و اندازه گیری داروی ریزاتریپتان از مایعات بیولوژیک

داروی ریزاتریپتان در درمان حملات حاد میگرن کاربرد دارد. این دارو در مقایسه با سایر داروهای ضد میگرن تاثیر سریع تری دارد. عوارض جانبی مصرف ریزاتریپتان شامل مشکلات قلبی و اسپاسم عروق کرونر، کاهش هوشیاری، خشکی دهان، مشکلات تنفسی و گوارشی می باشد. لذا با توجه اهمیت درمانی ریزاتریپتان و عوارض جانبی آن که موجب محدودیت دوز مصرفی دارو می گردد، مانیتورینگ و اندازه گیری داروی فوق در مایعات بیولوژیک بدن به منظور بررسی های فارماکوکینتیکی و کلینیکال امری لازم و اجتناب ناپذیر است که در طی مراحل درمان به نحوه تجویز و پایش دوز داروی مورد نظر کمک می کند. بدیهی است به منظور اندازه گیری مقادیر بسیار اندک ریزاتریپتان در مایعات بیولوژیک، می بایست از روشی استفاده نمود که قدرت شناسایی و تفکیک این دارو را داشته باشد. در این مطالعه با مقایسه انواع روش های میکرواستخراج، روش میکرو استخراج نوار حلال سه فازی با استفاده از فیبر تو خالی2 به همراه کروماتوگرافی مایع با عملکرد بالا3 جهت اندازه گیری داروی ریزاتریپتان در نمونه های بیولوژیک به منظور تعیین دوز داروی مورد نظر انتخاب گردید. روش فوق در مقایسه با سایر روش ها اقتصادی تر است و با صحت و دقت مناسب، می تواند گامی موثر در پیشبرد اهداف تامین سلامت جامعه بردارد.

rizatriptan is used to treat acute migraine attacks. this drug has a faster effect compared to other anti-migraine drugs. side effects of rizatriptan include heart problems and spasm of coronary arteries, loss of consciousness, dry mouth, respiratory and digestive problems. therefore, considering the therapeutic importance of rizatriptan and its side effects that limit the dosage of the drug, monitoring and measuring the above-mentioned drug in biological body fluids for the purpose of pharmacokinetic and clinical investigations is necessary and inevitable, which is necessary during the treatment stages according to the prescription method. and monitoring the dose of the desired drug helps. obviously, in order to measure very small amounts of rizatriptan in biological fluids, it is necessary to use a method that has the ability to identify and separate this drug. in this study, by comparing various microextraction methods, the three-phase solvent strip microextraction method using hollow fiber2 along with high-performance liquid chromatography3 was chosen to measure the drug rizatriptan in biological samples in order to determine the dose of the desired drug. the above method is more economical compared to other methods and with proper accuracy and accuracy, it can take an effective step in advancing the goals of providing health to the society.
