   reconstruction of mouse uterine tissue in vitro and in vivo using nanospun polycaprolacton/ gelatin/ polydimethylsiloxane hybrib scaffold

چکیده    Aim and background: uterus is the main organ of reproduction in women. healthy uterine endometrium plays an important role in implanting and supporting a fertilized egg. therefore, uterine endometrial repair and regeneration can cure some endometrial disorders that lead to infertility. methods: based on the structure of rat uterine tissue, a nanofiber scaffold were designed and fabricated by electrospinning. mouse uterine tissue cells were cultured on target scaffold. mtt test performed to evaluate cell viability on the scaffold. mouse embryos were cultured on the target scaffold and examined with immunofluorescents staining. the tubular scaffold was designed and constructed according to the structure of the rat uterine horn. the tubular scaffold replaced one of the horns of the rat uterus and was evaluated 30 days after the reconstruction surgery by hematoxylin & eosin staining and immunohistochemistry assay to evaluate the tissue formation. results and discussion: the growth and proliferation of mouse uterine cells on the target scaffold was well done. the mouse embryo was well adapted to the cellular scaffold and the tubular scaffold transplanted to the mouse uterus was covered h3by the proper tissue of the mouse endometrium. the designed tubular scaffold showed that the cell infiltration between the nanofibers is better than cell growth on the control flat surface.conclusion: we developed a hybrid scaffold can be a good support for implantation and growth of a fertilized egg and can be a good backup for the future whole uterine tissue engineering
