Aim and background: nanofiber-based scaffolds have been broadened by a broad spectrum of physicochemical properties such as high surface-to-mass ratio, the ability to be shaped in various forms, and resembling the features of native ecm that are the most challenging in the tissue engineering field. to succeed in scaffold implantation, the design and fabrication of suitable scaffolds are needed and further criteria should be considered; the ability to store bioactive molecules, preserve their stability and bioactivity, and controllable their release rate. so, emulsion electrospinning is a promising tool for the safe incorporation of agents into nanofibrous scaffolds that can be used for biomedical applications. methods: in the present study, the core-shell nanofibrous mat is composed of pcl as a core and silk fibroin-peo as a shell fabricated using an emulsion electrospinning technique. the biological and biomechanical properties of the construct were investigated. results and discussion: based on the result, it was found that the core-shell mat was fabricated correctly, demonstrating the core and shell separately. the sem result showed the bead-free structure of the nanofibers. the mechanical behavior of a core-shell was also higher than a shell. furthermore, it can be observed that excellent adhesion of cells on the fabricated core-shell mat. conclusion: our present study provides a new drug delivery system based on pcl/sf-peo core-shell nanofibrous that could dope with nanoparticles or drugs. we believe that the optimized core-shell scaffold is an ideal candidate for a drug delivery system
