Aim and background: docetaxel is a potent anticancer drug used to treat breast, and advanced non-small cell lung cancer. the marketed form of dtx (taxotere® ) were often subjected to hypersensitivity after administration. in order to avoid these disadvantages more study have been developed. the amphiphilic pdma-b-ppfpa used to load dtx due to its micelle formation. the physicochemical characteristics of dtx-loaded pms such as size, zeta potential, cmc, the release study and cytotoxicity were investigated. methods: polymeric micelles were prepared by film method. the polymer and dtx solution were mixed and evaporated to obtain dtx/copolymer matrix. hepes-sucrose (10mm) were used to hydrate. to remove the no-encapsulation drug centrifuged for in 14000 rpm for 10 min. finally, encapsulation efficacy was determined by hplc method. mtt test used for evaluate the cytotoxicity.results and discussion: dtx-pm were found to have size 38.50 nm and the zeta potential of −2.15 mv. dtx was well incorporated into pm with ee% 80. the dtx-pm released approximately 65% dtx during 48h. cytotoxicity test against4t1cell showed that dtx-micelles had better in vitro cytotoxicity compared taxotere as control. micelles internalized together with the entrapped dtx in the cytoplasm, probably via endocytic mechanism led improved cytotoxicity compared free drug.conclusion: dtx well incorporated into polymeric micelles with spherical shape showed that dtx-micelles had better in vitro cytotoxicity than taxotere
