مطالعه تشریحی و شمارش عدد کروموزومی گونه stipa capensis در ایران

Stipa capensisthun. گونه ای یکساله از جنس stipaمتعلق به خانواده گندمیان است. شواهد تشریحی بخش های رویشی گیاهان گلدار و داده های سیتولوژیکی برای توضیح مسائل طبقه بندی و تعیین حدود گونه ها از سده های پیش مورد استفاده قرار گرفته است. هدف از این مطالعه، شمارش کروموزومی و توصیف تشریحی ساقه و برگ این گونه و تعیین اهمیت کاربرد آن در رده بندی می باشد. در مطالعه پیش رو، پس از جمع‌آوری و شناسایی نمونه‌های گیاهی، از برگ و ساقه آنها به روش دستی برش عرضی تهیه و بهترین برش‌ها برای رنگ‌آمیزی و مطالعه با میکروسکوپ نوری انتخاب شدند.. جهت مشاهده کروموزوم ها، از سلول های مریستمی انتهای ریشه استفاده شد به این ترتیب که ریشه های بریده شده ابتدا توسط آلفابرومونفتالین پیش تیمار شده و سپس در محلول اتانول و اسید استیک تثبیت گردید و سپس رنگ آمیزی ریشه ها به کمک اورسئین 2 درصد انجام شد. در نهایت از گستره متافازی عکسبردای و کروموزوم ها شمارش گردید. در ساختار تشریحی برگ، وجود سلول های حبابی فن شکل، اپیدرم تحتانی باکوتیکول ضخیم، تعداد دستجات آوندی نامتقارن در دو سمت رگبرگ میانی مشاهده گردید که از جمله صفات متمایز کننده گونه s. capensis از سایر گونه های این جنس شناخته می شود. شمارش کروموزوم های این گونه با سطح پلوئیدی 36= n2، تایید کننده سایر مطالعات انجام شده است. بر اساس داده های موجود، داده های تشریحی گونه مذکور در ایران برای اولین بار گزارش می گردد.

stipa capensis thun. is an annual species of the stipa genus belonging to poaceae. anatomical evidence of the vegetative parts of flowering plants and cytological data have been used to explain the problems of classification and delimitation of species since centuries ago. the aim of this study is chromosomal counting and anatomical description of the stem and leaf of s.capensis and determining the importance of its use in classification. in the leading study, after collecting and identifying the plant samples, their leaf and stem cross section were cut manually and the best sections were selected for staining and studying with a light microscope. to observe the chromosomes, the meristem cells at the end of the root were used. it was done in this way that the cut roots were first pre-treated with alphabromonaphthalene and then fixed in ethanol and acetic acid solution, and then they were stained with the help of 2% orcein. finally, the metaphase area was photographed and chromosomes were counted. in the anatomical structure of the leaf, the presence of fan-shaped bubble cells, the lower epidermis with a thick cuticle, the number of asymmetric vascular bundles on both sides of the middle vein were observed, which are among the distinguishing features of s. capensis species from other species of this genus. chromosome counting of this species with a ploidy level of 2n=36 confirms other studies. based on the available data, the anatomical data of the mentioned species is reported for the first time in iran.
