مطالعه ارتباط ژن ms4a6a سیستم ایمنی با امکان بروز آلزایمر

بیماری آلزایمر،مهمترین اختلال عصبی و شایعترین آسیب عصبی است که خودش را با از دست دادن تدریجی حافظه و آگاهی به صورت پیشرونده نشان میدهد و در نهایت منجر به مرگ بیمار میشود.در این بیماری هم عوامل ژنتیکی و هم عوامل محیطی دخیل هستند و تا 79 درصد وراپت پذیری نشان میدهد.از جمله این ژنهای دخیل در ایجاد بیماری آلزایمر عبارتند از:app,psen1,psen2,ms4a,… .خانواده یms4a دارای خصوصیات ژنی ضعیفی میباشد ولی در حالت کلی نقشهایی برای اعضای این خانواده در نظر گرفته شده از جمله:پروتئینهایی بعنوان آداپتور،در سلولهای خونساز،در سیگنال دهی و تنظیم دقیق فعال سازی سلولها. ms4a6a از جمله اعضای این خانواده است که وراثت پذیری بالایی نشان میدهد، در چند مکان روی بازوی بلند کروزوموزم 11 قرار دارد.افزایش خطر ابتلا به بیماری آلزایمر (ad) قبلاً با یک snp به نام rs610932 در نزدیکی ژن ms4a6a مرتبط بوده است. هدف از این آزمایش تعیین کمیت بیان دو ایزوفرم ms4a6a در مغز افراد مبتلا به ad و غیر ad بود، به ویژه به عنوان تابعی از ژنوتیپ rs610932. طبق مقاله‌ای ms4a6a دارای چهار ایزوفرم مختلف است که گزارش شده است که در خون افراد مبتلا به ad در مقایسه با افراد غیر ad بیان متفاوتی دارند.دیده شده که در بیماری آلزایمر، دو ژن ms4a4a,ms4a6a افزایش بیان نشان داده اند. ولی به هر جهت،تحقیقات بیشتری نیاز است تا متوجه شویم چگونه این اتفاق رخ میدهد.

alzheimer s disease is the most important neurological disorder and the most common neurological damage that manifests itself with the gradual loss of memory and consciousness and eventually leads to the death of the patient. both genetic and environmental factors are involved in this disease and it shows 79% reproducibility. among these genes involved in the development of alzheimer s disease are: app, psen1, psen2, ms4a,…. the ms4a family has weak genetic characteristics, but in general, roles have been considered for the members of this family, including: proteins as adapters, in hematopoietic cells, in signaling and fine-tuning the activation of cells. ms4a6a is one of the members of this family that shows high heritability, located at several locations on the long arm of chromosome 11. increased risk of alzheimer s disease (ad) has previously been associated with a snp named rs610932 near the ms4a6a gene. the aim of this experiment was to quantify the expression of the two ms4a6a isoforms in the brains of ad and non-ad subjects, specifically as a function of rs610932 genotype. according to one article, ms4a6a has four different isoforms, which have been reported to be differentially expressed in the blood of ad subjects compared to non-ad subjects. it has been seen that two genes ms4a4a and ms4a6a have increased expression in alzheimer s disease. but in any case, more research is needed to understand how this happens.
