بررسی اثر سایتوتوکسیک عصارهی هیدروالکلی گیاه khorasanesis asparagus بر ردهی سلولی 231mb-mda و بیان ژنهای bax و 2b

از سرطانهای شایع در زنان سرطان پستان است و به دلیل عوارض جانبی روشهای درمانی شیمیایی، گیاهان که منابع مهم ترکیبات ضدسرطان هستند، میتوانند گزینه درمانی مناسب باشند. هدف از این مطالعه بررسی اثر سایتوتوکسیک عصارهی هیدروالکلی گیاه khorasanesis asparagus بر ردهی سلولی 231mb-mda و بیان ژنهای bax و 2bcl است. رده سلولی 231mb-mda و سلولهای نرمال -293hek از موسسه پاستور تهیه و کشت داده شد. سلولها به دو گروه تیمار و کنترل تقسیم شدند. عصاره گیاه khorasanesis asparagus روی گروه تیمار با غلظت به دست آمده از طریق 50ic (ml/μg24.5 ( تاثیر داده شد و سمیت آن با استفاده از assay mtt سنجیده شد. بیان 2 ژن bax و 2bcl با روش real pcr timeبررسی شد. rna از سلولهای گروه تیمار و کنترل استخراج و cdna ساخته شد. میزان بیان ژنها با استفاده از pcr time real و ct ∆∆مورد بررسی قرار گرفت. آنالیز آماری از طریق نرم افزار spss انجام شد. دادهها به صورت میانگین ± انحراف معیار )sd )بدست آمده از گروههای تحت تیمار بیان شد. اختالف بین گروهها بوسیله anova way-one و تست tukey آنالیز شده است. تاثیرعصاره khorasanensis asparagus در سلولهای سرطانی باعث کاهش زندهمانی آنها شده است و بیان ژنهای مرتبط با سرطان )2bcl و bax )را تغییر داده است که باعث کاهش بیان ژن 2bcl و افزایش بیان ژن bax شده است. با توجه به تاثیر آپوپتوزی عصاره khorasanesis asparagus بر رده سلولهای سرطانی 231mb-mda این عصاره در از بین بردن سلولهای سرطانی و تغییر بیان ژنهای مرتبط با سلولهای سرطانی تاثیر داشته در نتیجه این گیاه میتواند در درمان سرطان موثر واقع شود و به عنوان دارو در آینده مورد استفاده قرار گیرد

evaluation of cytotoxic effect of hydroalcoholic extract of asparagus khorasanensis on mda-mb231 cell line and expression of bax-bcl2 genes

one of the most common cancers in women is breast cancer, and due to the side effects of chemical treatment methods, plants, which are important sources of anti_x0002_cancer compounds, can be a suitable treatment option. the aim of this study is to investigate the cytotoxic effect of asparagus khorasanesis hydroalcoholic extract on mda-mb231 cell line and the expression of bax and bcl2 genes. mda-mb231 cell line and normal hek-293 cells were prepared and cultured from pasteur institute. the cells were divided into two treatment and control groups. the extract of asparagus khorasanesis affected the treatment group with the concentration obtained through ic50 (24.5 μg/ml) and its toxicity was measured using mtt assay. the expression of 2 genes bax and bcl2 was checked by real time pcr method. rna was extracted from the cells of the treatment and control groups and cdna was made. gene expression levels were analyzed using real time pcr and ∆∆ ct. statistical analysis was done through spss software. the data were expressed as mean ± standard deviation (sd) obtained from the treated groups. the difference between groups was analyzed by one-way anova and tukey s test. the effect of asparagus khorasanensis extract on cancer cells has decreased their survival and changed the expression of cancer-related genes (bcl2 and bax), which decreased bcl2 gene expression and increased bax gene expression. according to the apoptotic effect of asparagus khorasanesis extract on mda-mb231 cancer cell line, this extract has an effect in destroying cancer cells and changing the expression of genes related to cancer cells. it can be effective in the treatment of cancer and be used as medicine in the future.
