بررسی پتانسیل پروبیوتیکی باکتری­های اسیدلاکتیک جدا شده از محصولات لبنی سنتی و آنالیز ژنومی آنها در شمال ایران، گیلان

اهمیت باکتری های اسید لاکتیک در تعادل میکروبیوتا، عملکرد سیستم ایمنی، پیش گیری از بروز یا پیشرفت بیماری‌ها شامل سرطان اثبات شده است. هدف این مطالعه جداسازی، شناسایی و انتخاب بهترین سویه لاکتوباسیلوس با فعالیت پروبیوتیک از محصولات لبنی بومی استان گیلان بوده است. تعداد 30 جدایه از محصولات لبنی بومی مناطق روستایی استان گیلان با روش های میکروبیولوژی و بیوشیمیایی بررسی گردید. خصوصیات پروبیوتیک آنها و سپس فعالیت آنتاگونیستی عصاره خام سویه ها در مقابل پنج باکتری بیماری زا به روش انتشار در چاهک و آنالیز ژنومی آنها برای بررسی 16srrna به روش pcr تعیین گردید.چهار جدایه توانستند رشد ایزوله های بالینی استافیلوکوکوس اورئوس، شیگلا فلکسنری،سودوموناس آئروژینوزا و ایکلای پاتوژن ادراری را مهار کنند ولی اثرات مهاری معنی داری در رشد باسیلوس سرئوس نداشتند. درآنتی بیوگرام سویه ها به وانکومایسین و جنتامایسین مقاوم و به آمپی سیلین حساس بودند. تنها دو جدایه از سویه ها در pcr توانستند باند واضحی نشان دادند. گونه ثبت شده دلبروکیی زیرگونه بولگاریکوس بود. عصاره ها در مقایسه، فعالیت ضد میکروبی بهتری در شرایط خنثی نشده و بدون تیمار با کاتالاز نشان دادند.با توجه به نتایج این تحقیق و خصوصیات پروبیوتیک های جدا شده می توان از این سویه ها به دنبال بررسی بیشتر جهت پیشگیری و درمان عده ای از موارد بالینی استفاده نمود.

investigation of probiotic potential of lactic acid bacteria isolated from traditional dairy products and their genomic analysis in northern iran, gilan

the importance of lactic acid bacteria in microbiota balance, immune system function, and prevention of diseases including cancer has been proved. the aim of this study was to isolate, identify, and select the best lactobacillus strain with probiotic activity from indigenous dairy products in gilan province. a total of 30 varieties of domestic dairy products in the rural area of gilan were studied by microbiological and biochemical methods. their probiotic properties were evaluated and then the antagonistic activity of the crude extracts of five pathogenic bacterial strains was tested by the well diffusion method. their genomes were analyzed for 16srrna by pcr.four strains were able to inhibit the growth of clinical isolates of staphylococcus aureus, shigella flexneri, pseudomonas aeruginosa and uropathogenic e.coli, but had no significant inhibitory effect on the growth of bacillus cereus. the strains were resistant to vancomycin and gentamicin and sensitive to ampicillin in the antibiogram test.the two strains showed different bands in the pcr analysis. the recorded species was lactobacillus delbrueckii subsp. bulgaricus. the extracts showed better antimicrobial activity under neutral and catalase-free conditions. based on the results of this study and the properties of the isolated probiotics, these strains can be further investigated for the prevention and treatment of specific clinical conditions.
