isolation and identification of pseudomonas aeruginosa from patients with urinary tract infection

Pseudomonas aeruginosa is a saprophytic organism widespread in nature and is an important human pathogen. this study evaluated the frequency of pseudomonas spp. in patients with urinary tract infection using pcr method. fifteen urine samples were collected from patients with urinary tract infection hospitalized in shiraz hospital. the samples were cultured on blood agar, macconkey agar, and pseudomonas agar, and the isolates were assessed for pseudomonas spp. detection using a pcr test targeting the 16s rrna gene (618 bp). the isolates were then sequenced and subjected to the national center for biotechnology information (ncbi) and matched with the homologous pseudomonas spp. sequences available in blast. phylogenetic analysis was finally performed: of 15 samples, 2 (20%) isolates were detected pseudomonas spp. which were finally identified p. aeruginosa using phylogenetic analysis (figure 1). the sequences were also deposited in genbank database with accession numbers of or083631 and or083627. urinary tract infection related to p. aeruginosa pathogen shows importance of specific measures to control the infection especially in hospitalized patients.
