Today, sterilization using immunological methods is very common. among the various methods, immunity against gonadotropin hormone-releasing hormone (gnrh) has been the most attention and has reached the commercial stage. the aim of this study is to evaluate experimental anti-gnrh vaccine to abrogate sex hormone secretion in a male rat model. in the first trial, which aimed to determine the effective dose, 4 groups of male rats were randomly selected and immunized with doses of 50, 100, 200 and 400 µg of gnrh protein with 30% alum as an adjuvant. in the second stage, the goal was to determine the best percentage of alum as an adjuvant؛ based on the results of the first stage, the dose of 400 µg was selected and they were protected in 3 groups with different percentages of alum: 30%, 50% and 70%. in both tests, vaccination was administered intramuscularly in two doses with an interval of 21 days the first and second tests showed that the vaccinated group with 400 µg of gnrh and 30% alum was significantly different from other groups in terms of both antibody response and testosterone reduction (p<0.05) considering all the above-mentioned subjects in addition to availability and easier and cheaper way of preparation, we came to this conclusion that 400 µg of gnrh and 30% alum satisfactorily met most of our favorite criteria and could be used in immunocastration vaccine production purposes successfully.
