preliminary setup and study design on the impact of mitotempo on nlrp3 gene expression in leukocytes of breast cancer-induced mice

Nlrp3 gene expression plays a complex role in cancer, exhibiting both tumor-promoting and suppressing effects depending on the cancer type. it promotes the progression of breast cancer. inhibiting nlrp3 with drugs has been proven to decrease tumor growth and improve survival rates in mice. mito-tempo, a mitochondria-targeted antioxidant, exhibits potent antioxidant and anti-inflammatory effects by regulating nlrp3 gene expression under a variety of conditions. since there is no detailed information about its effects on nlrp3 gene expression in breast cancer, this study may serve as a guide for future immunotherapy approaches in mammary gland tumor. a number of 18 balb c mice with an average age of 4-6 weeks were purchased and after one week of acclimatization were subjected to mammary cancer induction. two days after tumor induction, mice were injected weekly in control, mito-tempo, vincristine, and mito-tempo +vincristine groups, and after each injection, the weight and general condition of the mice were measured. after one week from the fourth injection, the mice were weighed and re-evaluated, and blood samples were collected from the members of each group and analyzed for the expression of nlrp3 gene in leukocytes. amplification, melting, and natural log fold change of nlrp3 have shown significant differences among the groups, which should be further analyzed. the results suggest a successful setup for our future study on the exact effect of mitotempo on nlrp3 gene expression with and without chemotherapy. activation of nlrp3 inflammasomes promotes metastasis and angiogenesis and inhibits apoptosis and facilitates tumor progression. according to these results, decreased expression of this gene with mitotempo could provide a novel treatment approach in breast cancer.
