اثر عصاره هیدرو الکلی چویل بر فعالیت حرکتی، استرس و افسردگی در موش‌های ایسکمی

زمینه و هدف: ایسکمی در مغز موش موجب ایجاد آسیب‌های نورونی شدید و به دنبال آن موجب اختلالات استرس و افسردگی می‌گردد. عصاره گیاه چویل ترکیبات فنلی دارد و آنتی اکسیدان قوی است که نقش بسیار مهمی در حفاظت از سلول‌های عصبی ایفا می‌کند. هدف از این مطالعه بررسی اثر حفاظتی عصاره گیاه چویل بر استرس و افسردگی در مدل ایسکمی در موش‌های نر است.مواد و روش‌ها: در این تحقیق 50 سر موش صحرایی نژاد ویستار در محدوده وزنی 200 تا 250 گرم به طور تصادفی در 5 گروه، سالم (کنترل)، ایسکمی هیپوپرفیوژن بدون دریافت عصاره و سه گروه ایسکمی هیپوپرفیوژن، به مدت 14 روز، روزانه مقدار 100، 200 و 400 میلی گرم بر کیلو گرم عصاره چویل را به روش تجویز داخل معدی یا گاواژ دریافت نمودند. برای ایجاد ایسکمی شریان‌های کاروتید عمومی دو طرفه جدا شده، از دو قسمت مسدود و از بخش میانی قطع شدند. ارزیابی استرس و اضطراب در طی آزمون شنای اجباری و تست جعبه باز انجام گرفت.یافته ها: در موش‌های ایسکمی افزایش اضطراب و استرس یا کاهش فعالیت‌های حرکتی در طی آزمون شنای اجباری وتست جعبه باز مشاهده گردید. و تجویز دوزهای 100، 200 و400 میلی گرم در کیلوگرم عصاره گیاه چویل به ویژه، دوز400 و 200 منجر به بهبود نقایص ایجاد شده در استرس و افسردگی گردید (05/0>p).نتیجه گیری: عصاره چویل باعث کاهش اضطراب و افسردگی در مدل حیوانی ایسکمی شده است، که احتمالا به علت وجود بعضی از فنل ها و فعالیت آنتی اکسیدانی آن می‌باشد.

the effect of chuyil hydroalcoholic extract on motor activity, stress and depression in ischemic rats

background and purpose: ischemia in the brain of mice causes severe neuronal damage, followed by stress and depression disorders. chuvil plant extract contains phenolic compounds and is a strong antioxidant that plays a very important role in protecting nerve cells. the aim of this study is to investigate the protective effect of chuvil plant extract on stress and depression in ischemia model in male rats.materials and methods: in this research, 50 wistar rats weighing between 200 and 250 grams were randomly divided into 5 groups, healthy (control), hypoperfusion ischemia without extract and three hypoperfusion ischemia groups, for 14 days, 100 per day. they received 200 and 400 mg/kg of chuvil extract by intragastric administration or gavage. to create ischemia, bilateral common carotid arteries were isolated, blocked from two parts and cut from the middle part. assessment of stress and anxiety was done during forced swimming test and open box test.results: an increase in anxiety and stress or a decrease in movement activity was observed in ischemic rats during the forced swimming test and the open box test. and the administration of doses of 100, 200 and 400 mg/kg of chuvil plant extract, especially, doses of 400 and 200 led to the improvement of defects caused by stress and depression (p<0.05).conclusion: chuvil extract has reduced anxiety and depression in an animal model of ischemia, which is probably due to the presence of some phenols and its antioxidant activity.
