بررسی اثرات ژن کد کننده پپتید tachyplesin i خرچنگ نعل اسبی بر بیان ژن های bcl-2 و bax در سلول های achn

زمینه و هدف : سرطان در نتیجه تقسیم غیر قابل ‌کنترل سلول‌ها و در اثر عوامل محیطی و اختلال‌های ژنتیکی به وجود می‌آید. تومور مجموعه‌ای از سلول‌هاست که به دلیل رشد و تقسیم خارج از کنترل آن‌ها ایجاد می‌شود . هدف از انجام این پژوهش بررسی اثرات ژن کد کننده پپتید tachyplesin i بر بیان ژن های bcl-2 و bax در سلول های سرطان کلیه رده achn می باشد.مواد و روش ها: پلاسمید نوترکیب pcdna3.1(+)حاوی ژن tachyplesin i به روش شوک حرارتی وارد باکتری e.coli سویه top10 شد و سپس تخلیص شد . سپس وکتور نوترکیب حاوی ژن tachyplesin i به روش لیپوفکشن وارد سلول های achn شد و غربالگری توسط آنتی بیوتیک نئومایسین انجام گرفت. استخراج rna و سنتز cdna از سلول های ترانسفکت شده انجام شد. در نهایت واکنش real time rt-pcr به ‌منظور بررسی تغییر بیان ژن های bcl-2 و bax انجام شد.نتایج: پس از لیپوفکشن سلول ها در محیط کشت حاوی آنتی بیوتیک نئومایسین رشد کردند. سپس استخراج rna و سنتز cdna انجام شد. واکنش real time rt-pcr نشان داد که بیان ژن tachyplesin i در سلول‌های سرطان سینه رده achn سبب کاهش بیان ژن bcl-2 (p= 0.0198) و افزایش بیان ژن bax (p= 0.069) شد.نتیجه گیری : با توجه به این که حضور ژن tachyplesin i باعث افزایش معنی دار بیان در ژن bax و کاهش معنی دار بیان در ژن های bcl-2 می شود، احتمال می رود که بتواند سلول را به سمت آپوپتوز هدایت کند.

effects of tachyplesin i peptide coding gene on bcl-2 and bax gene expression in achn cells

background and aim: cancer occurs as a result of uncontrolled cell division and environmental factors and genetic disorders. a tumor is a collection of cells that develops due to their out-of-control growth and division. the aim of this study was to investigate the effects of the gene encoding bee venom melatonin on the expression of bcl-2 and bax genes in achn kidney cancer cells. materials and methods: recombinant plasmid pcdna3.1 (+) containing tachyplesin i gene was entered into top10 strain e. coli by heat shock and then purified. the recombinant vector containing the tachyplesin i gene was then inserted into achn cells by lipofection and screened with the antibiotic neomycin. rna extraction and cdna synthesis were performed from transfected cells. finally, the real-time rt-pcr reaction was performed to investigate the altered expression of bcl-2 and bax genes. results: after lipofection, the cells were grown in culture medium containing the antibiotic neomycin. then rna extraction and cdna synthesis were performed. real-time rt-pcr showed that the expression of tachyplesin i gene in achn breast cancer cells decreased the expression of bcl-2 gene (p = 0.0198) and increased the expression of bax gene (p = 0.069). conclusion: considering that the presence of tachyplesin i gene causes a significant increase in expression in bax gene and a significant decrease in expression in bcl-2 genes, it is likely that it can lead the cell to apoptosis.
