اثر سایتوتوکسیسیتی سوپرناتانت lacticaseibacillus paracasei بر رده سلولی سرطان روده بزرگ (ht-29)

مقدمه و هدف: سرطان روده یکی از مهمترین عوامل مرگ و میر انسان می باشد. به همین دلیل یافتن مواد جدید با منشا طبیعی از باکتریهای بومی احساس می شود. این مطالعه با هدف بررسی تاثیر سایتوتوکسی سوپرناتانت lacticaseibacillus paracasei lb12 بر رده سلولی سرطان روده بزرگ (ht-29) انجام شد.مواد و روش ها: ابتدا سویه lb12 l. paracasei در محیط mrs براث کشت داده شد. سپس در دور rpm 5000 به مدت 15دقیقه سانتریفیوژ شد و سوپرناتانت (مایع رویی) جداسازی و از فیلتر 22/0 میکرومتر عبور داده شد. در نهایت سوپرناتانت لیوفیلیزه شد. برای بررسی فعالیت سایتوتوکسیک، رده سلولی ht-29 در غلظتهای مختلف سوپرناتانت سویه l. paracasei در مدت زمانهای 24،48،72 تیمار شد و سپس میزان توان زیستی توسط تست mtt بررسی شد.یافته ها: تست mtt با گرمخانه گذاری 24 ،48 و 72 ساعته سلول های ht-29 با تیمار (سوپرناتانت سویه l. paracasei ) نشان داد که میزان توان زیستی وابسته به دوز و زمان است و با افزایش غلظت، میزان توان زیستی نسبت به گروه کنترل کاهش معنی دار نشان دادند (05 /0

cytotoxicity effect of lacticaseibacillus paracasei supernatant on colon cancer cell line (ht-29)

introduction and purpose: intestinal cancer is one of the most important causes of human mortality. for this reason, it is felt to find new materials with natural origin from native bacteria. this study was conducted with the aim of investigating the cytotoxic effect of lacticaseibacillus paracasei lb12 supernatant on colon cancer cell line (ht-29).materials and methods: first, l. paracasei strain lb12 was cultured in mrs broth medium. then it was centrifuged at 5000 rpm for 15 minutes and the supernatant was separated and passed through a 0.22 mm filter. finally, the supernatant was lyophilized. to investigate the cytotoxic activity, ht-29 cell line was treated with different concentrations of l. paracasei strain supernatant for 24, 48, 72 hours, and then the biological potency was checked by mtt test.findings: mtt test with 24, 48 and 72 hours incubation of ht-29 cells with treatment (l. paracasei lb12 supernatant) showed that the amount of biological potency is dependent on dose and time and with increasing concentration, the amount of biological potency compared to the control group showed a significant decrease (p<0.05). the greatest effect of the supernatant was related to the concentration of 20 mg/ml in 72 hours after the treatment of the cells.conclusion: the results obtained from this test indicate that l. paracasei metabolites can reduce the growth of colorectal adenocarcinoma cells in a dose- and time-dependent manner. so that this effect increases with the increase of treatment dose and time.
