شناسایی lncrnaهای اختصاصی سرطان کلورکتال بر اساس مطالعات بیوانفورماتیک و تایید آزمایشگاهی بیان آن ها در مدل سلولی

زمینه و هدف : سرطان شامل مجموعه ای از بیماری های ژنتیکی است که با رشد و تکثیر غیرقابل کنترل سلول ها و انتشار آن ها در بدن همراه است. هدف از این مطالعه مقایسه میزان بیان lncrna های ccat1و crnde در نمونه های توموری و نرمال افراد مبتلا به سرطان کلورکتال (crc) بود.مواد و روش ها: در ابتدا با استفاده از دیتابیس های آنلاین و مطالعات بیوانفورماتیک دو lncrna در سرطان کلورکتال کاندید شدند. سپس، سلول های رده سرطانی کلورکتال sw948 و نرمال hdf تهیه گردید و پس از کشت سلول و استخراج rna تام سلولی، cdna سنتز گردید و تغییر سطح بیان این دو lncrna در سلول های سرطانی و نرمال با استفاده از روش real time pcr مورد بررسی قرار گرفت. نتایج: مطالعات بیوانفورماتیک نشان داد که lncrna های ccat1 و crnde بیشترین تغییر بیان را در سرطان کلورکتال دارند و کاندید مطالعات مولکولی شدند. سپس تغییرات سطح بیان lncrna های کاندید شده در رده های سلولیsw948 در مقایسه با رده hdf به کمک روش rt-qpcr بررسی شد (p< 0.05). واکنش real time rt-pcr نشان داد که بیان هر دو (p=0.0272) lncrna crnde و lncrna (p=0.0033) ccat1 در سلول سرطانی نسب به نرمال افزایش بیان نشان داده است.نتیجه گیری : با توجه به نتایج بدست آمده در این مطالعه، می توان عنوان کرد این دو lncrna در سرطان کلورکتال افزایش بیان معنی داری را نشان می دهند و می توانند به عنوان بیومارکر در سرطان کلورکتال معرفی شوند.

identification of colorectal cancer-specific lncrnas based on bioinformatics studies and laboratory confirmation of their expression in cell models

background and objective: cancer includes a set of genetic diseases that are associated with the uncontrollable growth and proliferation of cells and their spread in the body. the aim of this study was to compare the expression levels of ccat1 and crnde lncrnas in tumor and normal samples of people with colorectal cancer (crc). materials and methods: at first, using online databases and bioinformatics studies, two lncrnas were candidates in colorectal cancer. then, sw948 and normal hdf colorectal cancer cells were prepared and after cell culture and total cell rna extraction, cdna was synthesized and the change in the expression level of these two lncrnas in cancer and normal cells using real time pcr method, was investigated. results: bioinformatics studies showed that lncrnas ccat1 and crnde have the most expression changes in colorectal cancer and became candidates for molecular studies. then, changes in the expression level of candidate lncrnas in sw948 cell lines compared to hdf line were investigated using rt-qpcr method (p<0.05). the real time rt-pcr reaction showed that the expression of both lncrna crnde (p=0.0272) and lncrna ccat1 (p=0.0033) increased in cancer cells compared to normal. conclusion: according to the results obtained in this study, it can be said that these two lncrnas show a significant increase in expression in colorectal cancer and can be introduced as biomarkers in colorectal cancer.
