Understanding genetic factors influencing susceptibility to covid-19 is crucial for managing the pandemic. the interferon- induced transmembrane protein 3 (ifitm3) gene plays a pivotal role in the innate immune response against viral infections. the rs12252 polymorphism within ifitm3 has been associated with varying susceptibility to viral pathogens. this study aimed to investigate the association between the ifitm3 rs12252 polymorphism and susceptibility to covid-19 infection. a case-control study was conducted involving 100 covid-19 patients and 50 healthy controls. genomic dna was extracted from peripheral blood samples, and the rs12252 variant was genotyped using rt-pcr with fluorescent-labeled taqman probes. genotype and allele frequencies were calculated, and statistical analyses were performed using chi- square test. hardy-weinberg equilibrium was assessed for both groups. genotype frequencies among patients and controls were consistent with hardy-weinberg equilibrium (p = 0.07 for patients, p = 0.526 for controls). a significant difference was observed in the distribution of aa, ag, and gg genotypes between covid-19 patients and controls (p < 0.05). specifically, the aa genotype (60%) was more prevalent among patients. allele frequency analysis revealed a highly significant difference in the frequencies of the a and g alleles between the two groups (p < 0.001), with the a allele being more common in patients than in controls (79.3%). the findings indicate a significant association between the ifitm3 rs12252 polymorphism and increased susceptibility to covid-19 infection. individuals carrying the aa genotype or the a allele may have a higher risk of contracting covid-19.
