   polymorphysm of dc-sign(cd209) promoter-336 in indonesian children with dengue infection

چکیده    Understanding the genetic basis of dengue infection could provide better insight into dengue pathogenesis, particularly immunogenetics. however, data on the association of the dc-sign polymorphism with the severity of dengue virus infection (dvi) in indonesian children are limited. this study is the first to explore the dvi immunogenetics in indonesian children. a cross-sectional study was conducted on children hospitalized at dr. soetomo general academic hospital, surabaya, indonesia, between 2013 and 2024. the children were categorized into dengue fever (df), dengue hemorrhagic fever (dhf), and dengue shock syndrome (dss). the polymorphisms of dc-sign (cd209) rs4804803 were analyzed using pcr-rflp with msci enzyme restriction from buffy coats and dried blood on filter papers. forty-seven children from east java province were enrolled and confirmed with dvi, consisting of 12 with df, 28 with dhf, and 7 with dss (2 died). significant differences were observed in age (p < 0.001, fisher’s exact test), dvi status (p < 0.001, fisher’s exact test), outcomes in df, dhf, and dss (p = 0.003, fisher’s exact test), and outcomes in leukopenia and thrombocytopenia (p < 0.008, p = 0.04, chi-square test, respectively). the cd209 rs4804803 polymorphism demonstrated the genotypes of aa (37/47, 79%), ag (8/47, 17%), and gg (2/47, 4%). the correlation between genotypes or alleles of cd209 rs4804803 with the severity of dvi, plasma leakage, thrombocytopenia, leukopenia, and bleeding manifestations was not significant in this study. this suggests that dc-sign may not be the primary receptor for the dengue virus.
