بررسی تاثیر جهش در پلی مورفیسم rs1255190244 در ژن apc بر بروز سرطان کلورکتال به روش tetra arms-pcr

مقدمه و هدف: جهش بر روی ژن پولیپوزیس کولی آدنوماتوز (adenomatous polyposis coli) می تواند باعث سرطان روده بزرگ شود. سرطان روده بزرگ به رشد سلول‌های سرطانی در کولون یا رکتوم گفته می‌شود. هدف از این مطالعه تجزیه و تحلیل جهش در نقطه ژنی rs1255190244 در ژن apc در بیماران مبتلا به سرطان روده بزرگ در استان تهران است. مواد و روش ها: از سال 1398 تا 1399، تعداد 100 نمونه به میزان 10-6 میلی لیتر به ازای هر بیمار مبتلا به سرطان روده بزرگ که به طور قطعی تشخیص داده شده بودند، جمع آوری شد. سپس dna هر نمونه استخراج گردید و پس از بررسی کیفیت dna به روش الکتروفورز، پرایمرهای موردنیاز طراحی شدند. در مرحله بعد با روش tetra arms pcr میزان پلی مورفیسم rs1255190244 در ژن apc بررسی گردید. درنهایت نتایج با نرم افزار spss تحلیل شد. سطح معناداری (0/003>p) درنظر گرفته شد. یافته ها: نتایج حاصل از این مطالعه نشان می دهد که بروز سرطان کولورکتال در نقطه ژنی rs1255190244 در گروه بیمار در مقایسه با گروه کنترل با p<0/003 به طور معنی داری با بروز سرطان روده بزرگ ارتباط دارد. نتیجه گیری: از نتایج این مطالعه نشان داد، می توان از پلی مورفیسم rs1255190244 به عنوان یک مارکر برای جلوگیری از سرطان روده بزرگ استفاده کرد، اما برای بررسی بیشتر بر روی شکل گیری های دیگر لازم است تحقیقات بیشتری انجام شود.

background and aims: mutations in the adenomatous polyposis coli gene can cause colon cancer. colorectal cancer is the growth of cancer cells in the colon or rectum. the aim of this study was to analyze mutations in the rs1255190244 gene point in the apc gene in patients with colorectal cancer in tehran province.materials and methods: from 2019 to 2020, 100 samples of 6-10 ml per patient were collected from patients with colorectal cancer who were definitively diagnosed. then dna of each sample was extracted and after checking the quality of the dna by electrophoresis, the required primers were designed. in the next step, the amount of rs1255190244 polymorphism in the apc gene was investigated by tetra arms pcr method. finally, the results of the analysis and significance level (p <0.003) were considered with spss software.results: the results of this study show that the incidence of colorectal cancer at the rs1255190244 gene point in the patient group compared with the control group with p0.003> is significantly associated with the incidence of colon cancer.conclusion: from the results of this study, it can be concluded that rs1255190244 polymorphism can be used as a marker to prevent colon cancer, but further research is needed to investigate several other formations.
