بررسی اثر سمیت عصاره‌های هیدروالکلی گیاه پونه و ﻣﺮزه تابستانی بر رده‎های سلولی سرطان سینه انسانی ( mcf7و mcf10)

مقدمه: سرطان پستان یکی از بدخیمی‌های مهم می‌باشد که از هر هشت زن، یک نفر را مبتلا می‌کند. با وجود روش‌های درمانی جدید، مرگ ناشی از سرطان هر سال رو به افزایش می‌باشد. گیاهان دارویی از جمله منابع مهم ترکیبات ضد سرطان هستند. در این راستا، مطالعه حاضر با هدف تعیین اثرات سمیت سلولی عصاره هیدروالکلی گیاه پونه و مرزه تابستانی بر رده‌های سلولی mcf7 و mcf10 (سرطان پستان انسانی) در شرایط آزمایشگاهی انجام شد. مواد و روش‌ها: مطالعه حاضر در آزمایشگاه دانشگاه آزاد بروجرد در سال 1399 انجام شد. پس از تهیه عصاره هیدروالکلی هر دو گیاه، اثر غلظت‌های مختلف 125،60،30،15،7،3 و 500 میکروگرم بر میلی‌لیتر گیاه پونه و مرزه تابستانی بر رده‌های سلولی تهیه شده از بخش تومور بانک انستیتو پاستور ایران به مدت 24، 28 و 72 ساعت مورد بررسی قرار گرفت. اثر سمیت سلولی با استفاده از روش mtt () ارزیابی شد. جذب نوری نیز توسط دستگاه الایزا در طول موج 570 نانومتر اندازه‌گیری گردید. در این مطالعه از روش‌های آماری آنالیز واریانس یک طرفه استفاده شد. شناسایی ترکیبات توسط دستگاه gc/ms (gas chromatography–mass spectrometry) صورت گرفت. یافته‌ها: بالاترین درصد مهار رده سلول‌های سرطانی mcf7 و mcf10 در عصاره هیدروالکلی گیاه مرزه تابستانی در غلظت 500 میکروگرم بر میلی‌لیتر به میزان 96.558 درصد و در گیاه پونه با همین غلظت معادل 88.547 درصد بود. نتیجه‌گیری: نتایج نشان دادند که عصاره هیدروالکلی برگ گیاه پونه و مرزه تابستانی، اثر سمیت سلولی قابل توجهی بر رده سلول‌های سرطانی داشته و منجر به مهار شدید رشد رده سلولی سرطانی شده‌ است.

Cytotoxicity of Hydroalcoholic Extracts of Mentha pulegium L. and Satureja hortensis L. on Human Breast Cancer Cell Lines (MCF7 and MCF10)

Introduction: Breast cancer is one of the most important neoplasms affecting one out of eight women. Despite new therapeutic methods, the breast cancer mortality rate is increasing annually. Medicinal plants are considered important sources of anticancer compounds. The present study was conducted to evaluate the cytotoxic effects of hydroalcoholic extract of Mentha Pulegitim L. and Satureja hortensis L. on MCF7 and MCF10 breast cancer cell lines in vitro. Materials and Methods: This study was performed in the laboratory of Boroujerd Azad University, Boroujerd, Iran, in 2020. After the preparation of hydroalcoholic extracts of Mentha Pulegitim L. and Satureja hortensis L., the effect of different concentrations of 3, 7, 15, 30, 60, 125, and 500 g/ml of both plants were examined on cell lines prepared at the tumor section of the Department of Cell Bank of Pasteur Institute of Iran, Tehran, Iran, for 24, 28, and 72 h. The effect of cytotoxicity was evaluated using an MTT assay. The light absorption was determined at 570 nm wavelengths using an enzymelinked immunosorbent assay reader. In this study, a oneway analysis of variance was utilized. The identification of the compounds was performed using the Gas ChromatographyMass Spectrometry method. Results: The highest percentages of inhibition of MCF7 and MCF10 breast cancer cell lines were found in the 500 μg/ml concentrations of the hydroalcoholic extracts of Satureja hortensis L. and Mentha Pulegitim L.at 96.558% and 88.547%, respectively. Conclusion: The results of the present study showed that the hydroalcoholic extracts of Satureja hortensis L. and Mentha Pulegitim L. leaves had significant cytotoxicity effects on the breast cancer cell line and prohibited their growth extensively.
