چکیده
|
در این پژوهش توانایی رنگبری رنگ آزوی بتا نفتول نارنجی توسط باکتری نمک دوست (halomonaz strain gb) و اثر فاکتور غلظت، ph و دما بر رنگبری بررسی میشود. با استفاده از ثابت نگهداشتن یک عامل و تغییر عوامل دیگر شرایط بهینه رنگبری، حاصل میشود. بالاترین درصد رنگبری 88% است که درغلظت ppm 40 و دمای c˚38 و 9=ph، پس از 6 روز گرماگذاری بهدست آمد. غلظت در محدوده وسیع 15ppm 125 بررسی شد و به همین دلیل توانایی باکتری در رنگبری در دو بخش غلظتهای کمتر و بیشتر از ppm 40 تحلیل شد. اثرگذارترین فاکتور، ph بود و با افزایش ph در محدوده 69، رنگبری بهطور چشمگیری افزایش یافت. فاکتور دما در محدوده 35 c˚40 تاثیر چندانی بر رنگبری نداشت. مقایسه طیفهای حاصل از hplc و uvvis قبل و بعد از رنگبری و همچنین مشاهده پدیده توتومری در طیفهای حاصل از ir موید رنگبری قابل توجه میباشد. بهعلت اینکه باکتری مذکور نمک دوست و تحملپذیر نمک است، در تمامی مراحل آزمایش (w/v) 5% کلریدسدیم در نظر گرفته شده است.

Study of Decolorization Ability of Orange Azo βNapHtol by Halomonaz Strain Gb bacteria and Optimization the Effects of Concentration, pH and Temperature

Abstract
|
Through this research, the ability of orange Azo βNapHtol’s decolorization by Halomonaz Strain Gb bacteria and the efect of conectration, PH adn temoeratureon decolorization studied. The optimal conditions for decolorization were obtained via changing the level of different factors while keeping one factor in each test constant, is assayed. The final result shows the maximum percentage of decolorization (88%) by 40 ppm concentration, 38˚C and pH = 9 after 6 days of process. As a result of testing in a wide range of concentration (15125 ppm), decolorization ability of the bacteria divided in two parts: examinations with concentration of less than 40 ppm and more than 40 ppm. The most effective factor was pH and increasing pH in a range of 610 caused a remarkable increase in decolorization. Temperature of 3540˚C did not too have much effect on the decolorization. Comparison of arising spectrums from HPLC and UVVis before and after decolorization and observing the totomery ph enomenon in arising spectrums from IR confirmsthe considerable decolorization. Because of halopHilic and halotolerant bacteria, the amount of %5 (W/V) Sodium Chloride was added during all the tests.
