   نقش سیستم دوپامینرژیک مرکزی در میانجی‌گری اثرات اشتهاآور نوروپپتید w در جوجه‌های گوشتی

چکیده    زمینه و هدف: سیستم دوپامینرژیک مرکزی نقشی حیاتی در تنظیم اشتها ایفا می‌کند، اما تعامل آن با نوروپپتید w (npw) در پرندگان هنوز به‌خوبی روشن نشده است. مطالعه حاضر با هدف بررسی نقش گیرنده‌های دوپامینرژیک در پُرخوری ناشی از تزریق مرکزی npw23 در جوجه‌های گوشتی طراحی و اجرا شد.روش‌ها: در این مطالعه، مجموعاً 220 قطعه جوجه گوشتی (راس 308) در پنج آزمایش شرکت داده شدند. در هر آزمایش، محلول کنترل، npw23، آنتاگونیست‌های گیرنده‌های دوپامینرژیک (d1: sch23390, d2: ami-193, d3: ngb2904, d4: l 741,742) یا نوروتوکسین دوپامینرژیک (6-ohda)، به ترتیب به‌طور مستقل یا به‌صورت ترکیبی با npw23. به‌صورت داخل‌مغزی (icv) تزریق شد. سپس مصرف غذایی تجمعی 30، 60 و 120 دقیقه پس از تزریق اندازه‌گیری شد.یافته‌ها: تزریق npw23 با دوز 3 نانومول به‌طور معناداری مصرف غذای جوجه‌ها را افزایش داد. تزریق همزمان آنتاگونیست گیرنده d1 با npw23 این اثر را تقویت کرد (p ≤ 0.05)، در حالی که تزریق ترکیبی آنتاگونیست‌های گیرنده d2، d3 و d4 با npw23 تاثیر معناداری بر هایپرفاژی ناشی از npw23 نداشت (p>0.05). همچنین، اگرچه تجویز 6-ohda به‌تنهایی تاثیری بر اشتهای جوجه‌ها نداشت، اما هنگامی که به‌طور همزمان با npw23 تزریق شد، هایپرفاژی ناشی از npw23 را به‌طور معناداری افزایش داد (p ≤ 0.05).نتیجه‌گیری: این یافته‌ها نشان می‌دهند که افزایش اشتهای ناشی از npw23 از طریق تعاملات با سیستم دوپامینرژیک مرکزی، به‌ویژه از طریق گیرنده‌های d1، در جوجه‌های گوشتی میانجی‌گری می‌شود.

   the role of the central dopaminergic system in mediating the appetitive effects of neuropeptide w in broiler chickens

Abstract    background and aim: the central dopaminergic system plays a crucial role in appetite regulation; however, its interaction with neuropeptide w (npw) in birds remains poorly understood. this study was designed and conducted to investigate the role of dopaminergic receptors in hyperphagia induced by central npw23 injection in broiler chicks.methods: a total of 220 broiler chicks (ross 308) were involved in five experiments. in each experiment, the control solution, npw23, dopaminergic receptor antagonists (d1: sch23390, d2: ami-193, d3: ngb2904, d4: l 741,742), or the dopaminergic neurotoxin (6-ohda) were administered intracerebroventricularly (icv) in sequence, either independently or in combination with npw23. subsequently, cumulative food intake was measured at 30, 60, and 120 minutes after injection.results: injection of npw23 at a dose of 3 nanomoles significantly increased food intake in the chicks (p ≤ 0.05). the concurrent injection of the d1 receptor antagonist with npw23 enhanced this effect (p ≤ 0.05), whereas combined injections of d2, d3, and d4 receptor antagonists with npw23 had no significant effect on npw23 induced hyperphagia (p>0.05). furthermore, although administration of 6-ohda alone did not affect the chicks’ appetite, its simultaneous injection with npw23 significantly increased hyperphagia induced by npw23 (p ≤ 0.05).conclusion: these findings indicate that the increase in appetite induced by npw23 is mediated through interactions with the central dopaminergic system, particularly via d1 receptors in broiler chicks.
