   اثرات رژیم حاد سم کوپریزون در القای رفتارهای شبه افسردگی در موش‌های c57bl/6

چکیده    زمینه و هدف: بیماری مالتیپل اسکلروزیس (ام اس) یک بیماری هتروژن دمیلینه‌کننده دستگاه عصبی مرکزی است که بیماران مبتلا به آن، طیف وسیعی از علایم حسی، حرکتی، شناختی و روحی روانی را تجربه می‌کنند. افسردگی به‌عنوان یکی از علایم خلقی و روانی است که با شیوع نسبتاً بالایی در بیماران مبتلا به ام اس مشاهده می‌شود. هدف از این مطالعه، بررسی رفتارهای شبه افسردگی در موش‌های مدل حاد بیماری ام اس القا شده با رژیم غذایی حاوی سم کوپریزون است. روش‌ها: در این مطالعه، 14 سر موش نر c57bl/6 به‌طور تصادفی به دو گروه به شرح زیر تقسیم بندی شدند: گروهی که به‌مدت 6 هفته رژیم غذایی معمولی را دریافت کردند (گروه کنترل) و گروهی که به مدت 6 هفته رژیم غذایی حاوی 2/0 درصد سم کوپریزون را دریافت کردند (گروه کوپریزون). علاوه بر سنجش وزن حیوانات، دو آزمون اختصاصی مرتبط با رفتارهای شبه افسردگی شامل آزمون شنای اجباری و آزمون آویزان کردن از دم به‌طور هفتگی از موش‌ها گرفته شد. در پایان هفته ششم مغز حیوانات جهت بررسی ناحیه قشر پیش پیشانی از نظر آسیب‌شناسی بافتی مورد بررسی قرار گرفت.  یافته ‌ها: ارزیابی‌های رفتاری انجام شده توسط آزمون شنای اجباری بین گروه‌های کوپریزون و کنترل تفاوت آماری معناداری را در هفته‌های پنجم و ششم نشان داد (0/05 >p ). اما نتایج آزمون آویزان کردم از دم تنها در پایان هفته ششم تفاوت آماری معناداری را بین دو گروه کوپریزون و کنترل نشان داد (0/05 >p ). نتایج آسیب‌شناسی بافتی نشان داد که به‌دنبال 6 هفته رژیم غذایی حاوی 0/2 درصد سم کوپریزون، دمیلینیشن معناداری در ناحیه قشر پیش پیشانی مغز رخ نداده است.    نتیجه‌گیری: رژیم غذایی حاوی سم کوپریزون به‌صورت حاد قادر است منجر به بروز رفتارهای شبه افسردگی در موش‌های c57bl/6 گردد اگرچه که مشاهده این رفتارها همراه با میلین زدایی چشمگیری در ناحیه قشر پیش پیشانی مغز حیوانات نبوده است.

   effects of acute cuprizone intoxication on inducing depressive- like behaviors in c57bl/6 mice

Abstract    background and aim: multiple sclerosis (ms) is a heterogeneous demyelinating disease of the central nervous system, in which patients experience a wide range of sensory, motor, cognitive, and psychological symptoms. depression as one of the neuropsychological symptoms is observed with relatively high prevalence in ms patients. this study aims to investigate depression like behaviors in acute ms model mice induced by a diet containing cuprizone neurotoxin.methods: in this study, 14 male c57bl/6 mice were randomly divided into two groups as follows: a group that received a normal diet for 6 weeks (control group) and a group that received a diet containing 2 0.0% received cuprizone poison (cuprizone group). in addition to measuring the weight of the animals, two specific tests related to depression like behaviors, including the forced swimming test and the tail suspension test, were taken weekly from the mice. at the end of the sixth week, the prefrontal cortex region of the brain tissue underwent histopathological evaluation.results: behavioral evaluations performed by the forced swimming test between the cuprizone and control groups showed a statistically significant difference in the fifth and sixth weeks (p < 0.05). however, the tail suspension test data showed a statistically significant difference between the two cuprizone and control groups only at the end of the sixth week (p < 0.05). histopathological evaluations showed that following six weeks of a diet containing 0.2% cuprizone toxin, significant demyelination did not occur in the brain’s prefrontal cortex.conclusion: the diet containing cuprizone neurotoxin can lead to depression like behaviors in c57bl/6 mice, although the observation of these behaviors was not accompanied by significant demyelination in the prefrontal cortex of the animals.
