چکیده    زمینه و هدف: مالتیپل اسکلروزیس (ام اس) یک بیماری دمیلینه کننده دستگاه عصبی مرکزی است که بیماران را درگیر علایم حسی، حرکتی و شناختی متعددی می‌کند. هدف از این مطالعه ارزیابی عملکرد حافظه، بررسی آسیب‌شناسی بافتی و سنجش سطح استرس اکسیداتیو در بافت هیپوکامپ موش‌های مدل ام اس است.روش‌ ها: در این مطالعه موش‌ها به دو گروه کنترل که رژیم غذایی نرمال را دریافت کردند (کنترل) و گروهی که به مدت 6 هفته رژیم غذایی حاوی 2/0 درصد کوپریزون را دریافت کردند (کوپریزون). پس از سنجش وزن حیوان و آزمون رفتاری حافظه، ناحیه هیپوکامپ از نظر آسیب شناسی بافتی و مارکرهای بیوشیمیایی جهت سنجش سطح پارامترهای استرس اکسیداتیو مورد ارزیابی قرار گرفت.یافته‌ ها: وزن موش‌ها پس از 6 هفته رژیم کوپریزون نسبت به گروه کنترل به‌ طرز معناداری کمتر بود (0/05 >p ). ارزیابی های رفتاری انجام شده با آزمون ماز y بین دو گروه تفاوت معناداری نداشت، اما ناحیه هیپوکامپ در گروه کوپریزون به میزان زیادی دچار دمیلینیشن شده بود. سنجش پارامترهای مرتبط با سطح استرس اکسیداتیو نشان داد که میزان لیپیدپروکسیداسیون در گروه کوپریزون نسبت به گروه کنترل افرایش معناداری داشته است (0/05 > p ). به ‌علاوه، سطح گلوتاتیون احیاء و فعالیت کاتالاز در گروه کوپریزون نسبت به گروه کنترل کاهش معناداری را نشان داد (0/05 >p ).نتیجه‌ گیری : القای دمیلینیشن حاد با کوپریزون احتمالاً از طریق ایجاد استرس اکسیداتیو در ناحیه هیپوکامپ توانسته است که منجر به دمیلینیشن در بافت  این ناحیه گردد درحالی‌که بر روی عملکرد حافظه حیوانات اثر معناداری نداشته است.

Abstract    background and aim: multiple sclerosis (ms) is a demyelinating disease of the central nervous system that affects patients with many sensory, motor, and cognitive symptoms. this study aims to evaluate memory function and histopathology and measure the oxidative stress level in the hippocampus of the ms model in mice.methods: mice were divided into two groups that received a normal diet (control) and one that received a diet containing 0.2% cuprizone (cuprizone) for six weeks. after measuring the animal’s weight and behavioral memory test, the hippocampus was evaluated in terms of histopathology and biochemical markers to measure the level of oxidative stress parameters.results: the weight gain of mice after 6 weeks of the cuprizone diet was significantly lower than the control group (p < 0.05). although behavioral evaluations performed with the y maze test did not have significant differences between the two groups, the hippocampus in the cuprizone group revealed obvious demyelination. measuring the parameters related to the level of oxidative stress showed that the amount of lipid peroxidation in the cuprizone group increased significantly compared to the control group (p < 0.05). in addition, the reduced glutathione and catalase activity levels in the cuprizone group showed a significant decrease compared to the control group (p < 0.05).conclusion: the induction of acute demyelination with cuprizone, probably through the creation of oxidative stress in the hippocampus, was able to lead to demyelination in the tissue of this region, while it had no significant effect on the memory performance of the animals.
