   بررسی نقش عملکرد اورکسین در پاسخ استرس

چکیده    اورکسین یا هیپوکرتین نوروپپتیدی است که در نورون های هیپوتالاموس جانبی تولید می ‌شود. این نورون ها انشعابات خود را به سرتاسر مغز ارسال می ‌کنند و در هر منطقه با توجه به فعالیت اصلی ناحیه، نقش تعدیل کنندگی اعمال می ‌کنند. دو نقش اولیه کشف شده اورکسین شامل دخالت در فرایندهای خواب و رفتار غذا خوردن است. یکی از نقش ‌های مهم اورکسین، تنظیم سیستم های زیست-عصب شناختی مرتبط با پاسخ استرسی است. پایانه نورون های اورکسینی و گیرنده ‌های آن در نواحی مختلف مغزی مرتبط با حافظه از جمله هیپوکامپ قرار گرفته ‌اند. استرس به‌ عنوان یک چالش هومئوستاتیک منجر به بدعملکردی فرایندهای یادگیری و حافظه می ‌شود. اورکسین در بروز علائم رفتاری و فیزیولوژیک استرس با تشدید فعالیت هسته مجاور بطنی هیپوتالاموس و افزایش ترشح هورمون آزادکننده کورتیکوتروپین اثر می ‌گذارد که آن هم به نوبه خود عملکرد نورون های اورکسینی را تقویت می ‌کند. این فعالسازی دوطرفه، نوعی مکانیسم بازخورد مثبت و شکلی از حافظه استرسی را در مدار خود بوجود می‌ آورد. مطالعات بیشتر برای فهم بهتر عملکرد اورکسین در سیستم عصبی مرکزی و محیطی ضروری است و این یافته ‌ها می ‌تواند کمک ارزشمندی برای درمان بیماری ها به ‌عنوان مثال بیماری های اضطرابی باشد. در این مقاله به مرور یافته ‌های مطالعات اخیر در رابطه با اورکسین و نقش آن در سیستم استرس می ‌پردازیم.

   Evaluation of the role of orexinergic function in stress response

Abstract    Orexin or Hypocretin is a neuropeptide, produced in the lateral hypothalamic neurons. The neurons send their projections throughout the brain and apply regionspecific modulatory functions. Two discovered primary functions of orexin are the involvement of orexin in food intake and sleep regulation. The regulation of neurobiological mechanisms involved in the stress response is one of the most important orexin functions. Orexin terminals and receptors are distributed in many memory and stressrelated areas such as the hippocampus. Stress, as a homeostatic challenge, leads to the malfunction of learning and memory processes. Orexin affects the behavioral and physiologic signs of stress by enhancing the activity of the paraventricular nucleus and secretion of corticotropinreleasing hormone, which in turn reinforces the function of orexin neurons, as a reciprocal circuitry. This mutual activation establishes a kind of positive feedback mechanism and a form of stress memory in the circuitry. Further studies are necessary for a better understanding of central and peripheral orexin functions and these results may help treat diseases especially anxiety disorders. We will review the recent findings regarding orexin and its role in stress response. lrm;
