   اثرات افزایشی لینالول بر خواب القاء شده با پنتوباربیتال: امکان دخیل بودن مسیر گابائرژیک

چکیده    زمینه و هدف: لینالول یک ترکیب منوترپنوئیدی و جزء اصلی اسانس برخی گیاهان معطر از جمله اسطوخودوس می ‌باشد. اثرات ضداضطراب و آرامبخش این ترکیب پیشتر گزارش شده است. در این مطالعه اثرات تجویز سیستمیک لینالول و مکانیسم احتمالی تاثیر آن بر خواب القاء شده با پنتوباربیتال در موش سوری مورد بررسی قرار گرفته است. روش ‌ها: در مرحله اول این مطالعه، 24 سر موش سوری در 4 گروه مجزا لینالول را در دوزهای 10، 50 و 100 میلی گرم بر کیلوگرم شصت دقیقه پیش از تجویز پنتوباربیتال دریافت نمودند. بر مبنای نتایج، 50 میلی ‌گرم بر کیلوگرم به ‌عنوان دوز موثر لینالول تعیین گردید. در مرحله دوم، برای تعیین مکانیسم دخیل در اثر خواب آوری لینالول از تجویز داروهای دیازپام به‌ عنوان آگونیست گیرنده های بنزودیازپینی، فلومازنیل به عنوان آنتاگونیست گیرنده های بنزودیازپینی و وی 100635 به عنوان آنتاگونیست گیرنده های سروتونینی استفاده شد. یافته‌ ها: مطالعه حاضر نشان داد که لینالول به‌ ویژه در دوز 50 میلی ‌گرم بر کیلوگرم به‌ صورت معنی‌ دار و وابسته به دوز خواب القاء شده با پنتوباربیتال را ازطریق افزایش مدت زمان خواب و کاهش طول مدت به خواب رفتن تحت تاثیر قرار می ‌دهد. تجویز همزمان دیازپام اثرات سینرژیستی نشان داد، منجر به تسریع به خواب رفتن و افزایش زمان خواب القاء شده با پنتوباربیتال شد، درحالیکه تجویز فلومازنیل منجر به معکوس شدن این اثرات شد. تجویز همزمان وی 100635 تاثیری بر اثرات افزایشی لینالول بر خواب القاء شده با پنتوباربیتال نشان نداد.نتیجه ‌گیری: لینالول احتمالا از طریق تقویت مسیرگابائرژیک خواب القاء شده با پنتوباربیتال را افزایش می دهد.

   Additive effect of linalool on pentobarbital-induced sleep: possible involvement of GABAergic pathway

Abstract    Background and Aim: Linalool is a monoterpenoid compound that is a major component of the essential oils of some aromatic plants, including lavender. The anxiolytic and sedative effects of this compound have been previously reported. In this study, the effect of systemic administration of linalool and its possible mechanism of action on pentobarbitalinduced sleep in mice was investigated.Methods: In the first phase of this study, twentyfour mice in 4 separate groups received linalool at the doses of 10, 50 and 100 mg/kg sixty minutes before pentobarbital administration. Based on the results of the first phase of the study, 50 mg/kg was determined as the effective dose of linalool. In the next step, for determination of the involved mechanism in linalool hypnotic activity, diazepam as a GABA benzodiazepine receptor agonist, flumazenil a GABA benzodiazepine receptor antagonist, and WAY100635 a serotonin receptor antagonist were used.Results: Linalool, especially at the dose of 50 mg/kg, significantly and dosedependently affected pentobarbitalinduced sleep by increasing sleep time and decreasing sleep latency. Coadministration of diazepam with linalool showed synergistic effects and increased sleep time; however, by flumazenil administration these effects were reversed. Administration of WAY100635 did not cause any effect on the increasing effect of linalool on pentobarbitalinduced sleep.Conclusion: Linalool may possibly be effective on pentobarbitalinduced sleep through the GABAergic pathway.
