چکیده
|
زمینه و هدف: مسیرهای عصبی داخل هیپوتالاموسی نوروپپتید y (npy)، اپیوئیدها و کیس پپتین از مهم ترین تنظیم کننده های فعالیت تولیدمثلی هستند. مرفین بیان ژن npy و کیس پپتین (kiss1) را کاهش می دهد. در حالی که کیس پپتین یا نالوکسون اثرات تحریکی بر بیان ژن npy اعمال می کنند. در این تحقیق نقش مسیر کیس پپتین/gpr54 در میانجیگری اثرات مرفین بر بیان ژن npy در هیپوتالاموس موش های صحرایی نر بررسی شد.روشها: چهل و پنج موش صحرایی نر ویستار به وزن 250-230 گرم در 9 گروه پنج تایی سالین، مرفین ( mg/kg5)، نالوکسون ( mg/kg2)، کیس پپتین ( nm1)، پپتید234 ( nm1)، نالوکسون/مرفین، کیس پپتین/مرفین، نالوکسون/کیس پپتین، و مرفین/پپتید234 دریافت کردند. نالوکسون و مرفین به صورت زیرپوستی و کیس پپتین و پپتید234 از طریق بطن سوم مغزی تزریق گردید. هیپوتالاموس نمونه ها جمع آوری شد. بیان نسبی ژن npy با روش sqrtpcr تعیین شد.یافته ها: مرفین سبب کاهش معنی دار و کیس پپتین سبب افزایش معنی دار بیان ژن npy در مقایسه با سالین شد. تزریق همزمان کیس پپتین/مرفین سبب افزایش معنی دار بیان ژن npy در مقایسه با مرفین و کاهش معنی دار بیان این ژن در مقایسه با کیس پپتین گردید. تزریق همزمان نالوکسون/کیس پپتین بیان ژن npy را در مقایسه با سالین، نالوکسون یا کیس پپتین افزایش داد که این افزایش در مقایسه با سالین و نالوکسون از نظر آماری معنی دار بود. تزریق همزمان مرفین/پپتید234 بیان ژن npy را در مقایسه با گروه سالین و مرفین کاهش داد که این کاهش در مقایسه با سالین از نظر آماری معنی دار بود.نتیجه گیری: علاوه بر کاهش فعالیت مسیر کیس پپتین /gpr54، احتمال دارد مسیرهای عصبی دیگری نیز در میانجیگری اثرات مهاری مرفین بر فعالیت نورون های npy نیز دخالت داشته باشند.

Role of kisspeptin/GPR54 pathway in mediating morphine's effects on hypothalamic NPY gene expression in male rats

Abstract
|
Background and aim: Intrahypothalamic neural pathways including neuropeptide Y (NPY), opioids and kisspeptin are among the most important regulators of reproductive function. Morphine decreases NPY and kisspeptin (Kiss1) gene expression, while kisspeptin or naloxone increases NPY gene expressions. In the present study the role of kisspeptin/GPR54 pathway was investigated in mediating morphine #39;s effects on hypothalamic NPY gene expression in male rats.Methods: Forty five male Wistar rats weighing 230250 g in 9 groups (in each group n = 5) received saline, morphine (5 mg/kg), naloxone (2 mg/kg), kisspeptin (1nM), peptide 234 (1 nM) or coinjection of naloxone/morphine, kisspeptin/morphine, kisspeptin/naloxone, and morphine/peptide234. Naloxone and morphine were injected subcutaneously, while kisspeptin and peptide 234 were injected into third cerebral ventricle. The hypothalamus of the animals was removed and relative NPY mRNA levels measured by RTPCR.Results: Morphine significantly decreased NPY gene expression, while kisspeptin significantly increased the expression. NPY gene expression was significantly increased by kisspeptin/morphine compared to saline. However, the expression was significantly decreased by kisspeptin/morphine compared to kisspeptin alone. The injection of kisspeptin/naloxone increased NPY gene expression compared to saline, naloxone or kisspeptin alone. This increase was statistically significant compared to saline or naloxone groups. Morphine/peptide234 decreased NPY gene expression compared to saline or morphine but the decrease was only statistically significant compared to saline.Conclusion: It seems that in addition kisspeptin/GPR54 pathway, other intrahypothalamic neural pathways are also involved in the inhibitory effects of morphine on NPY neurons activity.
