   لوکوس سرولئوس، نگاهی به علل آسیب‌پذیری

چکیده    سیستم‌های مونوآمینرژیک ساقه مغز از جمله سیستم لوکوس سرولئوس-نورآدرنرژیک به علت ارتباطات گسترده در بسیاری از پردازش‌های مرکزی نقش دارند و بروز هرگونه آسیب در این سیستم‌ها طیف وسیعی از اختلالات سیستم عصبی را به دنبال خواهد داشت. هسته لوکوس سرولئوس به عنوان منبع اصلی نورآدرنالین مغز پستانداران اولین سیستم مدولاتوری سلسله اعصاب مرکزی است که از نظر آناتومی و بیوشیمی توصیف و تبیین شده است. این هسته به واسطه رهایش نورآدرنالین، مجموعه‌ای از آثار تحریکی و مهاری را در نواحی هدف خود اعمال می‌کند. طی چند دهه اخیر آسیب هسته لوکوس سرولئوس به عنوان یکی از عوامل و ویژگی‌های مشترک برخی بیماری‌های سیستم عصبی از جمله بیماری‌های نورودژنراتیو مطرح شده است. به‌علاوه احتمال دارد بروز این بیماری‌ها، خود تظاهرات مختلفی از یک فرایند پاتوفیزیولوژیک مشترک یعنی آسیب هسته لوکوس سرولئوس باشد. توجه به نقش هسته لوکوس سرولئوس به عنوان یک عامل مداخله‌گر در پاتوبیولوژی این بیماری‌ها همچنین می‌تواند ما را به نقش این هسته به عنوان یک هدف بالقوه تشخیصی و درمانی به‌خصوص در بیماری‌هایی مانند پارکینسون و آلزایمر رهنمون شود. دلیل آسیب‌پذیری زیاد نورون‌های هسته لوکوس سرولئوس و نیز حساسیت آن ها به آسیب‌های دیگر نواحی مغز، دقیقاً معلوم نیست اما به نظر می‌رسد ویژگی‌های خاص آناتومیک و خونرسانی این هسته، فسفریلاسیون پروتئین تائو، نیاز به سطح بالای انرژی، آلودگی‌های محیطی و استرس از جمله عوامل موثر باشند که در این مقاله مروری به آن ها پرداخته می‌شود.

   The locus coeruleus, a look at the causes of vulnerability

Abstract    The monoaminergic systems of the brainstem, including the locus coeruleusnoradrenergic system, are involved in several central processing due to their extensive projections. Any damage to these systems will lead to a wide range of nervous system disorders. Locus coeruleus nucleus, as the main source of noradrenaline in the mammalian brain is the first CNS modulatory system, which has been described in terms of anatomy and biochemistry. This nucleus, through noradrenaline, exerts a series of stimulatory and inhibitory effects on its target areas. Over the past few decades, locus coeruleus damage has been identified as a reason and a common factor in some diseases of the nervous system, including neurodegenerative diseases. In addition, these diseases are likely to be different manifestations of a common pathophysiological process, namely locus coeruleus damage. Attention to the role of the locus coeruleus nucleus as an intervening factor in the pathobiology of these diseases can also lead us to the role of this nucleus as a potential diagnostic and therapeutic target, especially in diseases such as Parkinson rsquo;s and Alzheimer rsquo;s diseases. The reason for the high vulnerability of locus coeruleus neurons, as well as their sensitivity to damage compared to other areas of the brain are unclear but it seems that specific anatomical and blood supply features, tau protein phosphorylation, need to high energy levels, environmental pollutions, and stress are among the effective factors, which they will be described in this review article.
