   بررسی تاثیر نانو کپسول اریتروپویتین بر اضطراب ناشی از اختالل استرس پس از سانحه

چکیده    زمینه و هدف: فشار روانی پس از سانحه (ptsd)یکی از بیماری های اضطرابی است. ptsdباعث تغییر در بیان ژن های خانواده bcl2 و در نتیجه باعث بروز آپوپتوز در هیپوکمپ می ‌گردد. اریتروپویتین به عنوان یک فاکتور حفاظت عصبی می تواند از مرگ نورونی جلوگیری کند. اما این پروتئین توانایی عبور از سد خونی مغزی را ندارد. هدف مطالعه حاضر بررسی اثرات ضدآپوپتوز نانو کپسول اریتروپویتین در ptsd و مقایسه آن با اریتروپویتین است.روش ‌ها: با استفاده از مدل تک استرس طولانی مدت، ptsd در موش های صحرایی نر نژاد ویستار القاء شد. حیوانات بلافاصله پس از القاء، نانوکپسول اریتروپویتین و اریتروپویتین با دوز iu/kg 5000 را به ترتیب 1 و 3 بار به ‌صورت داخل صفاقی دریافت کردند. یک هفته بعد حیوانات مورد آزمون رفتاری، شمارش سلول های هیپوکامپ و بررسی سطوح بیان ژن های bcl2 و bax قرار گرفتند.یافته ‌ها: مطالعات رفتاری نشان داد نانوکپسول اریتروپویتین و اریتروپویتین، پاسخ های رفتاری اضطراب و ترس حساس شده را به طور معنی ‌داری نسبت به گروه کنترل کاهش (0/001 > p) دادند. همچنین کاهش تعداد سلول های هیپوکامپ ناشی از مرگ سلولی بهبود یافت. نتایج واکنش زنجیره ای پلیمراز نشان داد که ptsd در هیپوکامپ میزان بیان ژن bcl2 را کاهش و میزان بیان ژن bax را افزایش می دهد. تیمار دارویی این روند را معکوس نمود و به حالت طبیعی نزدیک کرد. نتیجه ‌گیری: هم اریتروپویتین و هم نانو کپسول اریتروپویتین دارای اثرات ضداضطراب و ضدآپوپتوزی در مدل حیوانی ptsd هستند. نانو کپسول اریتروپویتین در مقایسه با اریتروپویتین با دوز کمتری این اثرات را نشان می‌ دهد.

   Effect of erythropoietin nano-capsules on anxiety caused by post-traumatic stress disorder (PTSD)

Abstract    Background and aims: Posttraumatic stress disorder (PTSD) is one of the anxiety disorders. PTSD causes changes in the expression of the genes of the Bcl2 family in hippocampus and, as a result, causes apoptosis. Erythropoietin, as a neuroprotective agent, can prevent neuronal death in brain damage. But this protein has no ability to across the bloodbrain barrier. The aim of this study was to evaluate antiapoptotic effects of erythropoietin nanocapsules in PTSD and compare it with erythropoietin.Methods: PTSD was induced in male Wistar rats using the longterm single stress model (Singleprolonged stress). Immediately after induction, the animals received erythropoietin Nanocapsules and erythropoietin at a dose of 5000 IU/kg intraperitoneally 1 and 3 times/day, respectively. One week later, the animals underwent behavioral testing, hippocampal cell counts, and measuring Bcl2 and bax gene expression levels.Results: Erythropoietin Nanocapsules and erythropoietin significantly reduced the anxiety and fear compared to the PTSD group. Furthermore, drug therapy increased number of hippocampal cells, which were decreased by PTSD. PTSD decreased Bcl2 and increased bax hippocampal expression. However, these changes were reversed by drug treatment and reach the control level.Conclusion: Both erythropoietin and erythropoietin Nanocapsules show anxiolytic antiapoptotic effect in rat model of PTSD. Erythropoietin Nanocapsules showed these effects by lower doses than erythropoietin.
