چکیده
|
زمینه و هدف: از آنجا که فرآورده های استخراج شده از گیاهان دارویی نقش مهمی در درمان سرطان دارند، این مطالعه با هدف بررسی اثر سمیت عصاره های برگ تیمار شده بادرشبی با القاء کننده های مختلف بر روی چهار رده سلول سرطانی انجام شد.روش ها: پس از دو سال کشت بادرشبی در کرمان، محلول پاشی برگی با اسید سالیسیلیک و اسید بتا آمینوبوتیریک یک ماه قبل از برداشت در غلظت 0، 0/5 و 1 میلی مولار انجام شد. سپس اثر سمیت سلولی هر عصاره در دو غلظت 0/25 و 0/125 میلی گرم بر میلی لیتر بر روی 4 رده سلول سرطانی در 5 تکرار با روش mtt ارزیابی شد. . 3^10 × 3-5 سلول در پلیت های 96 خانه ای کشت شدند و درصد سمیت سلولی پس از 72 ساعت محاسبه گردید.یافته ها: از میان چهار رده سلول سرطانی انسانی، سلول های سرطان کبد و پستان بیشترین حساسیت را نسبت به عصاره های تام برگ تیمار شده بادرشبی بسته به تیمار انجام شده با مقادیر مختلف القاء کننده های اسید سالیسیلیک و اسید بتا آمینوبوتیریک در هر دو غلظت 0/25 و 0/125 میلی گرم بر میلی لیتر پس از گذشت 72 ساعت نشان دادند. نتایج نشان داد که کمترین حساسیت در برابر عصاره ها در هر دو غلظت، متعلق به سلول های سرطان کولون می باشد. کاربرد هر دو القاء کننده اسید سالیسیلیک و اسید بتا آمینوبوتیریک در شرایط رشد گیاه باعث افزایش درصد سمیت سلولی بر روی رده hep-g2 شد، در حالی که بر روی ردهht-29 اثر قابل ملاحظه ای نداشت. نتیجه گیری: عصاره های تیمار شده بادرشبی در غلظت بالا، سمیت سلولی قابل توجهی بر روی سلول های سرطانی کبد و پستان از خود نشان دادند. البته، مطالعات بیشتری نیاز است تا اجزای تشکیل دهنده عصاره جدا و اثرات آن ها مشخص شوند.

Abstract
|
Background and aims: Since products extracting from medicinal plants play an important role in the treatment of cancer, this study was performed to investigate the cytotoxicity effects of the total extracts obtained from the leaves of Dracocephalum moldavica (L.) treated with different elicitors on four human cancer cell lines.Methods: Following cultivating the Dracocephalum moldavica in Kerman for two years, foliar spraying was carried out using salicylic acid and betaaminobutyric acid at 0, 0.5 and 1 mM concentrations one month before harvesting. Then, the cytotoxic effect of each extract at two concentrations; 0.25 and 0.125 mg/ml was evaluated towards four human cancer cell lines at 5 replications using MTT assay. 35 × 103 number of the cells were seeded in 96well plates and the percentage of cytotoxicity was determined after 72 h of incubation.Results: Among the four cancer cell lines, liver and breast cancer cells demonstrated the most sensitivity against the total extracts obtaining from the treated leaves of D.moldavica depending on the type treatment with different amounts of salicylic acid and betaaminobutyric acid elicitors at two concentrations; 0.25 and 0.125 mg/ml after 72 hours of incubation. Results also showed that the lowest sensitivity to the extracts belonged to the colorectal carcinoma cells at two concentrations. Usage of both salicylic acid and betaaminobutyric acid as elicitors in the process of plant growth increased the percentage of cytotoxicity on HepG2 cell line, while it had no remarkable effect on HT29 cells.Conclusion: Total extracts obtaining from the treated leaves of D. moldavica at a high concentration displayed a considerable cytotoxicity towards human breast and hepatocellular carcinomas. Indeed, further studies are needed to separate the constituents of the extracts and investigate their effects.
