تاثیر کوتریماکسازول بر رونویسی قطعه تکراری 529 و فاکتور نکروز توموری آلفا در مغز موشهای آلوده به توکسوپلاسما گوندی

چکیده
|
زمینه و هدف: توکسوپلاسموز یک عفونت انگلی با شیوع جهانی است که بوسیله توکسوپلاسما گوندی ایجاد میشود. علائم عفونت حاد شبیه سرماخوردگی است ولی عفونت مزمن بدون علامت است. در این مطالعه اثر کوتریماکسازول بر تیتر انگل و پاسخ التهابی در مغز موش های مبتلا به توکسوپلاسموز بررسی شده است.روشها: موشهای سوری با تزریق صفاقی کیست انگل به توکسوپلاسموز مبتلا شدند. توالی 529rep (معرف بار انگل) و tnf alpha; (معیار پاسخ التهابی) در مغز موش ها با روش واکنش زنجیره ای پلیمراز کمی، در هفته های 1 تا 8 پس از تزریق کیست و نیز متعاقب درمان با کوتریموکسازول اندازه گیری شد. درمان با کوتریموکسازول ده روز پیش از شروع هفته چهارم و هشتم عفونت شروع شد و بمدت 10 روز ادامه یافت. یافتهها: از روز 4 تا 14پس از تزریق کیست، موش ها دچار آسیت شدید شدند. مقادیر 529rep نشان داد انگل هفته اول وارد مغز شده و هفته دوم و سوم با سرعت تکثیر مییابد. بیان tnf alpha; از هفته دوم شروع شده و به اوج می رسد و هفتههای بعد کاهش می یابد. کوتریموکسازول بطور معنیداری موجب کاهش معنی دار رونویسی 529rep و tnf alpha; در هفته 4 و 8 عفونت شد. درمان در هفته 4 با افزایش معنی دار وزن موشها (0/05 > p) نسبت به گروه درماننشده همراه بود.نتیجهگیری: نتایج این مطالعه نشان داد که در موش های سوری مبتلا به توکسوپلاسموز، تجویز مزمن کوتریموکسازول با مهار تکثیر انگل و پاسخ التهابی در مغز موشها و بهبود بیماری همراه است.

Effect of co-trimaxazole on REP-529 and TNF-α transcription levels in the brain of Toxoplasma gondii infected-mice

Abstract
|
Background and aims: Toxoplasmosis is a globally prevalent parasitic infection caused by Toxoplasma gondii. A flulike syndrome is seen in the acute phase of infection with no obvious symptoms in the chronic phase. In this study effect of cotrimaxazole on parasite load and inflammation in the brain of infected mice was examined.Methods: Mice were infected by intraperitoneal injection T. gondii cysts. Transcription of REP529 (as indicator of parasite load) and TNF alpha; (as indicator of inflammation) were measured in the mice brain by RTqPCR, at weeks 18 after injection of cysts, and after treatment with cotrimoxazole. Cotrimoxazole was administered 10 days before the weeks 4 and 8 of the infection for 10 days.Results: Severe ascites was observed during 414 days after injection of cysts. The REP529 transcription level indicates that the parasite enters the brain 1 week after cyst injection and proliferated rapidly in the second and third weeks. TNF alpha; level increased and reached the maximum level in the second week and then decreased in the next weeks. Cotrimoxazole significantly reduced transcription of both REP529 and TNF alpha; during 4 and 8 weeks of infection. Cotrimoxazole caused significant (p < 0.01) weight gain in mice at the week 4 of infection compared to the untreated group.Conclusion We found chronic administration of cotrimoxazole to mice infected by T. gondii can inhibit parasite proliferation and inflammation in the brain and cure the disease.
