تاثیر آمینوسیکلوپروپان کربوکسیلیک اسید و اسید سالیسیلیک بر ترمیم پذیری و بیان ژنهای Gai1 و Lox2 در جوانهزنی بذرهای زوال یافته سویا (Glycine Max)

چکیده
|
سویا یا glycine max (l.) merrill منبع اولیه روغن گیاهی است. بذرهای سویا معمولاً توانایی جوانهزنی را در طول ذخیرهسازی بلند مدت حتی در شریط مطلوب از دست میدهند. فاکتورهای زیادی در زوال بذر موثر هستند که شامل عوامل ژنتیکی، آسیب مکانیکی، رطوبت نسبی، دمای محیط ذخیرهسازی، محتوی رطوبت بذر، وجود میکروفلورا، رسیدگی بذر و عوامل دیگر میباشد که در صورت کاهش کیفیت بذر، شرایط را برای اهداف کشت نامناسب میکند. هدف از این آزمایش بررسی پاسخهای فیزیولوژیک بذر و همچنین شناسایی نقش ژنهای lox2 و gai1 در زوال بذر تحت تاثیر اتیلن و اسید سالیسیلیک در جوانهزنی بذر زوال یافته سویا بود.مواد و روشها: به منظور تعیین اثر زوال بر بذر سویا و همچنین تعیین اثر آمینوسیکلوپروپان کربوکسیلیک اسید (acc) و اسید سالیسیلیک بر بهبود زوال بذر، طی این تحقیق بذرهای سویا در معرض پیری تسریع شده به مدت صفر، 6 و 10 روز و پیری طبیعی به مدت 6 ماه قرار گرفتند. بذرها پس از پیری با اسید سالیسیلیک با غلظت 50 میکرو مولار و acc (پیش ماده اتیلن) با غلظت 10 میکرو مولار به مدت 6 ساعت در دمای 25 درجه سلسیوس تیمار شدند. همچنین دستهای بذر پس از آزمون پیری تسریع شده و طبیعی بدون هیچ تیماری به عنوان شاهد (بذر خشک) استفاده شدند. درصد جوانهزنی، میزان قند کل، گلوکز و فرکتوز بذر مورد بررسی قرار گرفت. همچنین بیان ژنهای gai1 و lox2 در بذرهای خشک و همچنین طی 6 و 12 ساعت تحت اثر آب، اسید سالیسیلیک و acc به روش q-rt- pcr بررسی شد.یافتهها: نتایج جوانهزنی نشان داد که با افزایش تعداد روزهای پیری، جوانهزنی کاهش مییابد. میزان قند کل در بذرهای 6 روز پیر تفاوت معنیداری با بذر پیر نشده نداشت؛ اما میزان قند کل در بذور 10 روز پیر نسبت به بذرهای پیر نشده افزایش معنیدار داشت. با افزایش سطوح پیری تسریع شده از صفر به 10 روز، محتوی گلوکز و فروکتوز در بذرهای خشک افزایش یافت. ژنها هم بیان متفاوتی را در روزها و ساعتهای مختلف از خود نشان دادند. با افزایش پیری از صفر به 10 روز، بیان gai1 افزایش یافت. همچنین بیان lox2 طی پیری تسریع شده از صفر روز به 6 روز افزایش یافت. بیان lox2 در بذرهای پیر طبیعی خشک نسبت به بذرهای پیر نشده افزایش یافت. هورمون اسید سالیسیلیک و acc اثرات متفاوتی را بر موارد اندازهگیری شده داشتند.نتیجهگیری: در مجموع میتوان چنین استنباط کرد که زوال بذر و کاهش بنیه نتیجه برآیند فرآیندهای تخریبی متعدد و اختلال در فعالیتهای فیزیولوژیکی بذر است. این پژوهش نشان داد که پیری با افزایش در میزان قندکل، گلوکز و فرکتوز همراه میباشد. همچنین بیان ژنهای دخیل در مسیر جوانهزنی نیز دستخوش تغییر میشوند. افزایش بیان ژن lox2 در هر دو مسیر پیری تسریع شده و پیری طبیعی مشاهده شد. بیان gai1 در پیری تسریع شده افزایش و در پیری طبیعی کاهش یافت.

The Effect of ACC and Salicylic Acid on Germination and GAI1 and LOX2 Genes Expression in Deteriorated Soybean Seeds (Glycine max)

DOR: 98.1000/23831251.1398. 6.61.12.2.1578.41Extended AbstractIntroduction: Soybean (Glycine max (L.) Merrill) is the primary source of vegetable oil. Even in desirable conditions, soybean seeds lose their viability in long term storage. Many factors contribute to seed deterioration, including genetic factors, mechanical damage, relative humidity, storage temperature, seed moisture content, existence of microflora, and seed maturity, which reduce seed quality and make seeds unfit for cultivation purposes.Materials and Methods: In order to investigate the effects of seed deterioration on seed germination and also the effects of salicylic acid and ethylene on the improvement of deteriorated seeds of G. max., accelerated aging test for 0, 6 and 10 days and natural aging test for 6 months were conducted. After aging conditions, seeds were imbibed with 50 micro;M salicylic acid and 10 micro;M ACC (precursor of ethylene) for 6 hours at 25 °C. In addition, after natural and accelerated aging tests, a bunch of seeds was used without any hormonal treatment (i.e., dry seeds) as control seeds. The seeds rsquo; germination percentage, total sugar, fructose, and glucose were investigated. Moreover, the gene expression of GAI1 and LOX1 was measured on dry seeds and under imbibition of water, salicylic acid and ACC at 6, 12 hours using QRTPCR method.Results: The germination results showed that increasing number of aging days led to a decrease in germination. Total sugar content in seeds aged for 6 days did not have a significant difference, as compared with nonaged seeds. However, total sugar content in seeds aged for 10 days was significantly higher than nonaged seeds. Increasing accelerated aging levels from 0 days to 10 days led to increases in glucose and fructose contents in dry seeds. In addition, genes exhibited different expressions in different days and hours. Increasing aging from 0 days to 10 days led to increases in GAI1 gene expression. Moreover, LOX2 expression increased in accelerated aging from 0 to 6 days. LOX2 gene expression in naturally dried aged seeds also increased and was higher than that in nonaged seeds. SA and ACC had different effects on measured values.Conclusion: In general, it can be concluded that the deterioration of seed quality and vigor result from numerous degradation processes and disruption in seeds rsquo; physiological activity. This study showed that aging is associated with an increase in total sugar, glucose and fructose levels. In addition, the expression of the genes involved in the germination is also affected. Increases in LOX2 gene expression were observed in both accelerated aging and natural aging pathways. GAI1 gene expression increased in accelerated aging. However, in normal aging, it decreased. Highlights:Identifying the role of LOX2 and GAL1 genes in soybean seed deterioration.Investigating seeds rsquo; physiological responses under natural and laboratory aging conditions.
